电针对帕金森病干扰素-γ、肿瘤坏死因子-α和白介素-1β影响

【摘要】 目的:静止性颤、肌肉僵直、行动徐缓和体位不稳仍是帕金森病(Parkinson’s di sease,PD)的主要症状。当前,存在多种假说来解释PD的发病机制,既引起多巴胺细胞退行性改变的病因机制仍不十分清楚[1],目前,神经免疫引起极大的关注[2]。临床上,我们观察到针灸治疗能十分明显地减轻和缓解静止性震颤及肌肉僵直。因此,我们想探讨针灸可能的神经免疫神经保护作用。方法:在脑定位仪下,把6-羟基多巴,注入左侧纹状体。连续2周针灸治疗,1/日,2周后,用酶联检测法(ELISA),测定血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和干扰素-γ(IFN-γ)。结果:大鼠帕金森后,TNF-α、IL-1β、IFN-γ含量升高。有显著性差异。美多巴组、针灸组、美多巴+针灸组均能降低TNF-α、IL-1β、IFN-γ含量。其中,针灸组差异显著性最大。结论:在6-羟基多巴毁损纹状体模型中,针灸治疗显著降低TNF-α、IL-1β、IFN-γ含量,也许针灸参与神经炎症机制的调节.
【Abstract】 Objective The characteristic symptoms of Parkinson’s disease(PD) consisting of akinesia, rigidity and tremor.The aetiology of the neuronal death in Parkinson’s disease remains, until today,unclear.Several possible mechanisms for the degeneration of dopaminergic neurons are still elusive.Considerable evidence suggests that neuroinflammation and oxidative stress might play important roles in the pathogenesis of Parkinson’s disease.In the clinic,acupuncture is observed significantly to reduce and relaxe tremor.Therefore,we investigated the possible neuro protective effect of acupuncture to reveal the mechanism of neuroinflammation in the rat of Parkinson’s disease.Methods the rats received a unilateral 6-hydroxydopamine(6-OHDA) injection into the left striatum.Testing was carried out 2 weeks post-lesioning after acupuncture intervention. The expression of tumor necrosis factor-α(TNF-α)、IL-1βand IFN-γwas evaluated by ABC-ELISA method.Results acupuncture -treatment,significantly attenuated the expression of TNF-α、IL-1βand IFN-γ.Conclusions In 6-OHDA lesion striatum model,for PD,acupuncture-treatment signif- icant attenuated the expression of TNF-α、IL-1βand IFN-γ.Maybe acupuncture take part in adjust of mechanism of neuroinflammation in Parkinson’s disease.
