【摘要】 目的:复制大鼠动脉粥样硬化模型的同时用通塞脉片(TSM)干预,观察TSM抑制大鼠动脉粥样硬化形成的作用。方法:雄性Wistar大鼠,随机分为5组,每组10只,为正常组、模型组、辛伐他汀组、TSM高剂量组和TSM低剂量组。除空白对照组给以普通饲料喂养外,其他组饲以高脂饲料及维生素D3,建立食饵性动脉粥样硬化模型。14周后检测血脂、超氧化物歧化酶(SOD)、丙二醛(MDA)及基质金属蛋白酶-9(MMP-9)、组织金属蛋白酶抑制因子-1(TIMP-1)。结果:模型组总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、动脉硬化指数(AI)、MDA含量均明显升高,SOD活性降低,MMP-9和TIMP-1表达均增强,TSM可降低TC、LDL-C、MDA含量和AI,升高SOD活性,抑制MMP-9、TIMP-1表达(P<0.05～0.01)。结论:TSM可以通过调节血脂代谢,抑制MMP-9的表达,阻止动脉粥样硬化的形成。更多还原

【Abstract】 Object:TO observe the protect effects of TSM on atherosclerosis model of rats.Methods: 50 wistar male rats were randomly divided into 5 groups:normal group(treated with 0.5% CMC-Na).model group(treated with 0.5%CMC-Na),simvastatin group(treated with SimvAStatin of 0.003g/kg·d),TSM high-dose group(treated with TSM of 12.44g/kg.d) and TSM low-dose group(treated with TSM of 6.22g/kg.d).Atherosclerosis model was established by feeding rats with high fat diet and administration with VD3 while normal control rats were_fed with standard chow.The five groups were administrated for 14 weeks.The serum levels of Triglyceride(TG), Total cholesterol(TC),High density lipoprotein(HDL),Low density lipoprotein(LDL),and plasm levels of superoxide dismutase(SOD),Maleic dialdehyde(MDA),and the expression of MMP-9/TIMP-1 on aorta were examined by immunohistochemistry.Reasults:Serum levels of TC、TG and LDL were decreased obviously while HDL was increased by treatment of TSM; release of MDA and synthesis of SOD were regulated by TSM in rats.Expression of MMP-9 and TIMP-1 on cells of aorta pectoralis were depressed to weak or no expression by TSM. Conclusions:TSM has good therapeutic activity on atherosclerosis in rats by regulating the disturbance of blood fat,enhancing the ability of SOD, reducing the release of MDA and inhibiting the expression of MMP-9 and TIMP-1 in vivo.更多还原
