不同品种新疆甘草液相色谱指纹图谱研究

【摘要】 目的研究甘草甲醇-水提取物中物质组成情况,对不同品种甘草进行HPLC指纹图谱分析。方法测定了新疆产三个药典规定品种野生甘草的指纹图谱,对其中甘草苷、甘草酸进行了含量测定;利用SPSS统计软件筛选了可区别品种的特征峰,并据此对三个品种进行了判别和聚类。结果对少量样品的初步判别和聚类取得了较为满意的结果。结论本法与前报甘草酸、甘草苷的定量、甘草液相色谱-质谱指纹图谱定性相结合,可直接了解各种甘草品种与化学成分变化的关系,探索了指纹图谱研究中应用统计工具的具体方法。
【Abstract】 Three widely used wild medicinal licorice plants,Glycyrrhiza Uralensis Fisch., Glycyrrhiza Glabra L.and Glycyrrhiza Inflata Bat which collected from Xinjiang were analyzed by RP-HPLC fingerprint.The 67%methanol extracts of licorice plants were gradient eluted by a mixture of water(with 0.1%H3PO4 for UV and 1%formic acid for ESI-MS detection, respectively)(solution A) and acetonitrile(solution B) on an ODS column.The eluents were detected by UV,DAD or ESI-MS detectors.The stepwise gradient profile of elution was 15%B at flow rate of 0.5mL/min(0min)-25%B,0.8mL/min(10min)-32%B(20min)-80%B(70min).The detection wavelengths were 275 nm(from 0 min),360 nm(from 19 min) and 248 nm(from 24 min).The canonical discriminance and the hierarchical cluster analysis for speciese showed a satisfying result.
