补肾方对流产大鼠胎盘胰岛素样生长因子介导孕激素生成的影响

【摘要】 目的:探讨补肾方对流产大鼠胎盘胰岛素样生长因子Ⅰ、Ⅱ(IGF-Ⅰ、IGF-Ⅱ)及其受体在胎盘滋养层细胞表达的影响及其与孕激素(P)分泌的关系。方法:溴隐亭皮内注射法建立流产大鼠模型,于孕1-8、1-11天灌服补肾方药,黄体酮肌注作为阳性对照组,采用放免法测定大鼠血清P水平;逆转录聚合酶链反应(RT-PCR)技术检测IGF-Ⅰ、IGF-Ⅱ及其受体在不同组动物胎盘中的mRNA表达。结果:溴隐亭模型组IGF-Ⅰ、IGF-Ⅱ、IGF-ⅡRmRNA表达较正常对照组明显降低;中药组及黄体酮组降低IGF-ⅠRmRNA表达,不同程度提高模型大鼠胎盘IGF-Ⅰ、IGF-Ⅱ、IGF-ⅡRmRNA表达。表达强度随孕天和给药时间逐渐升高,与血P有显著相关性。结论:补肾方益气清热、固肾安胎,能调控胎盘滋养细胞IGF-Ⅰ、IGF-Ⅱ及其受体的基因表达,并且调节滋养层细胞的增殖活性,促进孕激素合成分泌增加,从而达到保胎的目的。更多还原

【Abstract】 Abjective:To explore the influence of expression of IGF -Ⅰ、IGF -Ⅱand their receptors gene respectively on rat abortion placenta with replenishing kidney formula,the same time to assay relationship with progesterone secretion.Methods:The model rats of Bromocriptine sc during 6 - 8days of pregnancy induced early abortion was established,adopting respectively herbs in high dosage、herbs in low dosage、progesterone affect model rat,after 9、12 days,progesterone levels in the serum were measured by radioimmunoassay.Simultaneity,semiquantitative reverse transcriptase polymerase chain reaction(RT -PCR) was applied to determine the expression of IGF -Ⅰ、IGF -ⅠR、IGF -Ⅱ、IGF -ⅡRmRNA of rat placenta.Results:Expression of IGF -Ⅰ、IGF -Ⅱ、IGF -ⅡRmRNA of Bromocriptine model group decreased significantly compared with those of the control group.Herbs group and progesterone group decreased IGF -ⅠR gene expression,increased the expression of IGF -Ⅰ、IGF -Ⅱ、IGF -ⅡRmRNA on different level,and strength increased gradually companied with the process of pregnancy and time of treating,there was obvious relativity with progesterone secretion in serum.Conclusion:Replenish kidney formula could invigorating Qi and clearing heat,reinforcing the kidney to prevent abortion.Regulated the expression of IGFs gene of placenta and synthesis of progesterone and trophoblast cell proliferative activity.To get to the aim of prevent abortion.更多还原
