生脉成骨胶囊对血管内皮细胞分泌ET-1、NO及PGl2功能的影响

【摘要】 目的探讨生脉成骨胶囊含药血清对人脐静脉内皮细胞株(ECV304)分泌内皮素1(ET-1)、前列环素I2(PGI2)和一氧化氮(NO)功能的影响。方法用ELISA法检测生脉成骨胶囊含药血清作用1h和2h后ECV304细胞培养上清中ET-1和PGI2浓度,用酶法检测细胞培养上清中NO浓度。结果生脉成骨胶囊含药血清作用1h后,高剂量组上清中ET-1浓度显著降低(P<0.05),低、中、高剂量组上清中PGI2浓度均显著降低(P<0.05),高剂量组上清中NO浓度显著升高(P<0.05)。刺激2h后,高剂量组上清中ET-1浓度升高,低、中、高剂量组上清中PGI2浓度与对照组相比,仍较低,但低剂量组上清中PGI2浓度与中剂量组和高剂量组相比,有显著性升高(P<0.05),高剂量组上清中NO浓度仍显著升高(P<0.05)。结论生脉成骨胶囊含药血清可抑制ECV304细胞分泌PGI2,促进其分泌NO,高剂量组含药血清表现为先抑制后促进ET-1的分泌,提示生脉成骨胶囊可能从多方面影响血管内皮细胞血管活性物质的分泌功能。

【Abstract】 Objective To study the effect of rabbits’ serum containing Shengmai Chenggu Capsule(SMCGC) on cultured ECV304 cell for its secreting function of ET-1,PGI2 and NO.Methods The concentration of ET-1,PGI(?) in cultured liquid of ECV304 cell was measured with ELISA method,The concentration of NO in the cultured liquid was detected with enzymic method.Results After being treated with SMCGC-containing rabbits’ serum For 1 hour and 2 hours,the concentration of ET—1 in high-dose-rabbit-serum cultured liquid of ECV304 cell significantly decreased(P<0.05),the concentration of PGI2 in low-dose-group,medium-dose-group and high-dose-group was significantly decreased(P<0.05),the concentration of NO in high-dosc-group increased(P<0.05),2 hours later,the concentration of ET-1 in high-dose-group significantly increased(P<0.05).Compared with control group,the concentration of PGI(?) in low-dose-group,medium-dose-group and high-dose-group was still at low level,but the concentration of PGI2 in low-dose-group increased significantly compared with that of medium-dose-group and high-dose-group(P<0.05), the concentration of NO in high-dose-group increased(P<0.05).Conclusion SMCGC has the function of inhibiting PGI(?) secreting and promoting NO secreting of ECV304 cell, as to the function of secreting ET-1,SMCGC has the effect of stimulating at the beginning and inhibiting later,this study shows SMCGC plays an important role in the secreting of vasoactive substances of ECV304 cell.
