青藤碱脂质体的处方优化及制备工艺研究
Study on the optimization of the formulation and preparation process of sinomenine liposomes

【摘要】 目的优化青藤碱脂质体的膜材处方,并进行制备工艺研究。方法采用乙醚注入法制备脂质体,以包封率为指标,采用混料均匀设计法优化其膜材处方,确定豆磷脂、胆固醇、维生素E等组分的用量。并在此基础上采用单因素考察法进行制备工艺研究,包括缓冲液用量、制备温度、超声时间等。结果最佳膜材处方为:青藤碱:豆磷脂:胆固醇:维生素E=8.92:60.35:28.81:1.91,缓冲液用量为50mL/g膜材,制备温度为50℃,超声时间为20min。结论按此优化处方及工艺制备的脂质体可获得满意的形态及包封率。
【Abstract】 Objective To optimize the formulation and preparation process of sinomenine liposomes.Methods By means of aether injection,mixture uniform design was adopted to screen the formulation of sinomenine liposomes to confirm the proportion of phospholipids,cholesterol and Vitamin E by the index of entrapment efficiency.And the Single-factor research was used to study the preparation process of the liposomes,including the volume of buffer solution,the preparation temperature and the ultrasonic time.Results The optimized formulation was sinomenine:phospholipids:cholesterol:vitamin E = 8.92:60.35:28.81:1.91.The volume of buffer solution was 50mL/g membrane,the preparation temperature was 50℃,and the ultrasonic time was 20 min. Conclusion Satisfactory shape and entrapment efficiency of the liposomes can be obtained by the optimized formulation and preparation process.
