绿茶多酚对实验性酒精性肝病大鼠肝脏脂质过氧化损伤及超微结构变化的影响研究

【摘要】 目的研究茶多酚及没食子酸表没食子儿茶素(EGCG)对实验性酒精性肝病大鼠肝脏脂质过氧化损伤及超微结构变化的影响。方法将48只大鼠随机分为正常对照组、模型组、茶多酚治疗Ⅰ组、Ⅱ组、EGCG治疗Ⅰ组、Ⅱ组,每组8只大鼠。以酒精+0.5ml鱼油灌胃制作酒精性肝病模型。茶多酚治疗Ⅰ、Ⅱ组另外分别给予200mg/kg、100mg/kg茶多酚灌胃治疗,EGCGⅠ、Ⅱ治疗组则分别给予100mg/kg、50mg/kg EGCG灌胃治疗。五周后处死大鼠,检测大鼠肝功能、超氧化物歧化酶(SOD)、脂质过氧化产物丙二醛(MDA)水平及肝脏超微结构变化。结果模型组大鼠血清ALT及肝组织SOD、MDA水平与正常对照组相比,显著升高(P<0.01),电镜示肝细胞内线粒体肿胀,部分嵴退化,粗面内质网扩张,滑面内质网增多呈空泡状,胞质内可见许多大小不等的脂滴。茶多酚及EGCG高低剂量治疗组均显著降低血清ALT及肝组织SOD、MDA水平,与正常组相比P>0.05,电镜示肝细胞胞质内均有脂滴,四个治疗组肝细胞胞质内均有脂滴,线粒体未见明显肿胀,粗面内质网平行排列,无明显滑面内质网。结论茶多酚及EGCG显著改善了酒精性肝病大鼠肝脏的超微结构变化,其机制与其抑制了酒精对大鼠肝细胞的脂质过氧化损伤有关。

【Abstract】 Objective:To investigate the effects of tea polythenols and Epigallocatechin -3-gallate(EGCG) on lipid peroxidation and ultrastructural changes in alcohol-induced liver injury in rats.Methods:SD rats were randomly divided into six groups:normal,model,two doses tea polyphenols treatment and two dose EGCG treatment groups.Alcoholic liver injury was induced by gavage of alcohol plus 0.5ml fish oil for 5 weeks.Tea polyphenols and EGCG treatment groups were administered with tea polyphenols 200 mg/kg,100 mg/kg,and EGCG 100mg/kg、50mg/kg respectively. Serum ALT activity,superoxide dismutase(SOD) activity and malondialdehyde(MDA) levels in the liver were detected,and liver ultrastructural changes were also examined.Results:Model group rats had a marked increase in levels of ALT,SOD,MDA,compared with normal group(P<0.01).Electron microscopy showed mitochondrial swelling,part mitochondrial cristae degeneration,expanded rough endoplasmic reticulum,increased smooth endoplasmic reticulum with the vacuolar shape,and abundant fat drops in cytoplasm in the liver in model group.A significant decrease in serum ALT activity,SOD activity and MDA levels in the liver was observed in tea polyphenols and EGCG treatment groups,all of which were comparable to model group.Hepatic ultrastructural changes caused by alcohol were markedly improved,with few mitochondrial swelling,almost normal endoplasmic reticulum after tea polyphenols and EGCG treatment;however the fat drops were still present in treatment groups. Conclusions:Tea polyphenols and EGCG can improve hepatic ultrastructural changes induced by alcohol,and the mechanism may involve the suppression of lipid peroxidation in the liver.
