茶多酚及EGCG对实验性酒精性肝病大鼠肝脏的保护作用及机制研究

【摘要】 目的研究茶多酚及EGCG对实验性酒精性肝病大鼠肝脏的保护作用及其作用机制。方法将48只大鼠随机分为正常对照组、模型组、茶多酚治疗Ⅰ组、茶多酚治疗Ⅱ组、EGCG治疗Ⅰ组、EGCG治疗Ⅱ组。以酒精+0.5ml鱼油灌胃制作酒精性肝病模型。茶多酚治疗Ⅰ、Ⅱ组另外分别给予200mg/kg、100mg/kg茶多酚灌胃治疗,EGCGⅠ、Ⅱ治疗组则给予100mg/kg、50mg/kg EGCG灌胃治疗。四周后处死大鼠,观察各组大鼠肝脏病理变化,并测定肝功能。ELISA检测血清TNF-α、IL-1、IL-18水平。结果模型组大鼠血清ALT水平与正常对照组相比,显著升高,并且肝组织表现肝细胞中度脂肪变性,点状坏死,炎性细胞浸润。茶多酚及EGCG治疗组显著降低血清ALT水平,与正常组相接近,肝组织仍可见肝细胞脂肪变性,但未见坏死及炎性细胞浸润。模型组血清TNF-α、IL-1、IL-18水平与正常组相比显著升高,但给予茶多酚及EGCG治疗后,其水平均明显下降。结论茶多酚及EGCG可减轻酒精性肝损伤的炎症与坏死,其机制可能与降低炎症细胞因子TNF-α、IL-1、IL-18等有关。
【Abstract】 Objective To study the effects of tea polyphenols and EGCG on alcohol-induced liver disease,and to explore its mechanisms.Methods Forty eight Wister rats were randomly divided into six groups:normal group,model group,two tea polyphenols treatment groups,and two EGCG treatment groups.Alcoholic liver injury was induced by gavage of alcohol plus 0.5ml fish oil for 4 weeks.Tea polyphenols treatment group rats were administered with 200 mg/kg、100 mg/kg tea polyphenols respectively;EGCG treatment groups with 100mg/kg、50mg/kg EGCG.Liver injuries were assessed by histopathologic examination and serum ALT levels.TNF-α,IL-1 and IL-18 serum levels were assayed by ELISA.Results Model group rats had marked increase in ALT levels compared with normal group, and their liver tissue showed hepatocytes fatty degeneration,focal necrosis and inflammatory cells infiltration.However,tea polyphenols and EGCG treatments resulted in decreased ALT levels and improved pathological changes.Serum TNF-α,IL-1 and IL-18 levels were elevated significantly in model group compared with normal group,but markedly suppressed by tea polyphenols and EGCG treatments.Conclusions Tea polyphenols and EGCG prevent inflammation and necrosis in alcohol-induced liver disease,and its mechanism may involve the inhibition of inflammatory cytokines, such asTNF-α,IL-1 and IL-18.
