人参皂苷Rgl对Aβ25-35诱导的星形胶质细胞核因子κB活化及炎症反应的影响

【摘要】 [目的]探讨人参皂苷Rg1对β淀粉样蛋白25-35(Aβ25-35)诱导的SD大鼠星形胶质细胞核因子-κB(NF-κB)的活化及其对炎性因子白细胞介素-6(IL-6)的影响。[方法]用人参皂苷Rg1预处理星形胶质细胞24 h(终浓度分别为0、2、4、8、16μmol/L),然后再用Aβ25-35和人参皂苷Rg1对星形胶质细胞共同孵育24 h,四甲基偶氮唑盐(MTT)检测各组的细胞活性;用激光扫描共聚焦显微镜(LSCM)检测分析各组的细胞浆与细胞核内NF-κB的激活程度;用酶联免疫吸附试验(ELISA)的方法检测IL-6的蛋白含量。[结果]人参皂苷Rg1各组的细胞活性均明显高于模型组(P<0.05),并且能下调NF-κB的激活水平,减少IL-6的分泌(P<0.05),该作用以2、4μmol/L为显著。[结论]人参皂苷Rg1对Aβ25-35诱导星形胶质细胞模型的神经保护作用可能是通过阻断NF-κB的活化,减弱炎症反应,从而提高了细胞的生存率,且此作用以低浓度为好。

【Abstract】 [Objective]To explore the effect of panaxsaponin Rg1 on nuclear factorκB activation and inflammation reaction of SD rats astrocyte induced by Aβ25-35.[Methods]To carry out pretreatment astrocyte for twenty-four hours by using panaxsaponin Rg1 and incubate in common astrocyte for twenty-four hours by using panaxsaponin Rgl and Aβ25-35.To determine cell activity of every group by MTT method,analysze activation extent of cytoplasm and intranuclear NF-κB by LSCM method,determine IL-6 protein content by ELISA method.[Result]The cell activity of panaxsaponin Rg1 group was obviously high than that of model group(P<0.05) and panaxsaponin Rg1 may down regulate activation level of NF-κB and reduce IL-6 level(P<0.05).[Conclusion]The panaxsaponin Rg1 had nerves protective effect on astrocyte induced by Aβ25-35. The effcts may be relationship with intercepting NF-κB activation,weakening inflammatory reaction and enhancing cellular survival rate and it had best effect on low density group.
