健脑益智冲剂对去皮层血管大鼠胶质纤维酸性蛋白、铁离子及一氧化氮合酶的影响

【摘要】 [目的]从扶正与祛邪的角度,探讨健脑益智冲剂治疗血管性痴呆的机制。[方法]采用去皮层血管法造成血管性痴呆大鼠模型,观察造模两周后大鼠胶质纤维酸性蛋白(GFAP)、铁离子及一氧化氮合酶(NOS)的改变及用药后的影响。[结果]去皮层血管术后,大鼠脑组织出现GFAP阳性星形胶质细胞;鼠脑中神经细胞NOS阳性反应减弱,一氧化氮(NO)生成减少;损伤周区脑铁离子含量升高。用药治疗后,鼠脑内GFAP阳性星形胶质细胞较模型组胞体增大,突起增多,长度增加均非常显著;神经细胞NOS阳性反应增强,NO生成增加;铁离子染色也呈阳性反应,但深棕色颗粒明显减少。表明健脑益智冲剂有促进反应性星形胶质细胞的增生、增加NOS神经元生成NO的量以及减轻脑内铁离子积累的作用。[结论]健脑益智冲剂促进反应性星形胶质细胞的增生,提高其吞噬、清除、调节、解毒保护及神经营养的作用,减少铁离子积累,防止脑内氧自由基的产生,减轻脑损伤。调节NO生成而改善脑功能。
【Abstract】 [Objective]To discuss effects and curative mechanisms of Jiannao Yizhi Decotion on treating vascular dementia in viewpoints of fuzheng and quxie.[Methods]The rat model of vascular dementia was established by the cortical devascularization operation.Changes in the expression of glial fibrillary acidic protein(GFAP),NOS positive neurons and accumulation of Fe2+ in the ischemic areas were investigated after operation or Jiannao Yizhi Decotion intervention.[Result]Expression of GFAP in reactive asrocytes was increased while NOS positive neurons reduced and Fe2+ accumulated 2 weeks after cortical devascularization.Jiannao Yizhi Decotion treatment promoted GFAP and NOS positive neuron expression,decreases Fe2+ deposits. [Conclusion]Jiannao Yizhi Decotion protected neuron from ischemic injuries by facilitated function of GFAP astrocytes include free radical or Fe2+ scavenging,antioxidant defense and secrete growth factors and improved effect of NOS positive neurons in learning and memorizing.
