Based on Quantum dots detection and immuno-fluorescence imaging of anti-nuclear antibody in different tissue cells

【摘要】 <正>Objective:To evaluate the effectiveness of Quantum dots(QDs) labeled antibody probe in the applications of indirect immunofluorescence assay(IIFA).Methods:IIFA was used for ANA detection based on QD655-tags,assay results which generated were also compared with that of FITC tags.Results:There existed results discrepancies at a certain degree between QD655- and FITC-tags,including centromere pattern,nuclear homogeneous pattern,nucleolar pattern and cytoplasmic granular pattern,etc,which generated by the target antigen positioned in different parts of the cells.QD655-tags was priority with cytoplasm antigen/antibody reaction,but weakly stained with interphase-cell nuclei,nuclear centromere,dsDNA,nucleosome and histone nuclear-antigens relatively.Yet,ANA results generated by QD655 and FITC fluorescent labellings were roughly approached conformity in split-phase cells.Conclusion:Based on QD-tags,IIFA used for ANA detection is feasible,and has advantages of sensitivity and stability. Synchronously,QD- and FITC-tags together used for ANA will achieve the better effectiveness.
