16S rRNA在肉毒梭菌分型鉴定中的应用

【摘要】 目的:建立一种快速,可靠的对肉毒梭菌进行分型鉴定的手段。方法:从肉毒梭菌中提取基因组DNA为模板,通过16S rRNA特异性引物进行PCR,扩增得到16S rDNA序列并测序。将结果通过Clustal和Mega两个软件分析并构建进化树分析其种属特异性。结果:与传统方法分型鉴定得到的结果一致。结论:16S rRNA在肉毒梭菌分型鉴定中具有快速,准确的优势,随着核糖体库的不断完善,有望成为细菌分型鉴定的标准依据。
【Abstract】 Objective:To establish a quickly and accurate method for detecting Clostridium botulinum by 16S rRNA. Methods:The Clostridium botulinum Genome was extracted and the 16S rRNA specific primers of Clostridium botulinum were designed and 16S rDNA genes were amplificated with PCR.Then the phylogenetic trees were constructed by Clustal and Mega.Results:Both 16S rRNA and conventional method have the same detecting results.Conclusion:16S rRNA has obvious advantage in speed and accuracy.
