微波加热法制备CdTe量子点及其在细胞标记中的应用

【摘要】 采用微波加热的方法,通过优化前驱体溶液中CdCl2、巯基乙酸、KTeH4的比例,改变前驱体溶液的pH值和微波加热的温度及时间,快速合成了水溶性CdTe/CdS核/壳结构的量子点。利用紫外-可见吸收光谱、荧光发射光谱、透射电子显微镜、X-射线光谱表征了量子点的光学特性、粒径和结构。利用该量子点作为荧光探针标记了狗肾细胞(MDCK),并用流式细胞仪对用于标记细胞的量子点的浓度进行了选择,用荧光显微镜对标记后的细胞进行了表征。结果表明,该探针具有较好的细胞相容性,在生物标记方面具有良好的应用前景。
【Abstract】 CdTe-CdS core-shell quantum dots were synthesized in aqueous solution by microwave irradiation.The key synthesis parameters(molar ratio of reagents,pH values,reaction temperature,and time) were fully discussed,and the optimum conditions were illuminated in this paper.Absorption and fluorescence spectra showed that these prepared CdTe quantum dots had good optical properties.The diameter and structure were characterized by the transmission electron microscopy(TEM )and X-Ray photoelectron spectra. The MDCK was labeled by the CdTe fluorescence probes successfully.Concentrations of CdTe quantum dots for labeling were selected by flow cytometry and the labeled cells were characterized by fluorescence microscope.The results showed that the fluorescence probe have good biocompatibility and will be promised in the application for biologic labeling.
