大气细颗粒物对BALB/c小鼠心血管系统的免疫损伤机制研究

【摘要】 大气细颗粒物与心血管疾病密切相关,但其作用机制尚未阐明,本研究探索Thl7/Treg细胞在大气细颗粒物致心血管系统免疫损伤和炎症反应中的作用机制。采集大气细颗粒物,采用气管滴注的方法对BALB/c小鼠进行染毒,染毒剂量为10 mg/kg.bw,对照组气管滴注生理盐水,染毒24 h后,取心肌和脾脏组织。检测心肌组织中与Thl7/Treg细胞亚群相关的细胞因子及转录因子的表达水平,同时分别测定脾脏组织中CD4+、CD4+Foxp3+和CD4+IL-17+细胞的含量,分析各细胞亚群占CD4+细胞的比例。结果表明,大气细颗粒物能引起CD4+IL-17+和CD4+Foxp3+细胞明显增加。同时,心肌中IL-17,TGF-β1和Foxp3的mRNA表达明显增加。结果表明Thl7/Treg细胞不平衡可能是大气细颗粒物致心血管系统免疫损伤的作用机制。

【Abstract】 Ambient fine particles closely associated with cardiovascular disease.However,the mechanism is not elucidated yet.The aim of this study is to explore the mechanism of Thl7/Treg in fine particle-induced immune and inflammatory injury of cardiovascular system.BALB/c mice were exposed by intratracheal instillation to fine particles with the doses of 0 and 10 mg/kg b.w, respectively.Twenty-four hours after the exposure,the rats were killed and spleen and heart were collected.Cardiac histology demonstrated exacerbated cardiac lesions in BALB/c mice.Reverse transcriptase polymerase chain reaction(RT-PCR) assay assessed the mRNA expression of interleukin-17A,transforming growth factor-beta 1(TGF-β1) and Foxp3.Meanwhile,the CD4+ IL-17+ and CD4+Foxp3+ were measured in spleen using flow cytometry technique.The results showed that the levels of CD4+IL-17+ and CD4+Foxp3+ increased in mice after exposed to fine particles. Compared to control,the mRNA expression of IL-17A,TGF-β1and Foxp3 increased in fine particles exposure group.The results indicated that imbalance of Thl7/Treg might be one of the mechanisms of cardiovascular injury induced by fine particles.
