铅对小鼠细胞DNA损伤作用和增殖变化的研究

【摘要】 目的了解铅对小鼠细胞DNA损伤作用和增殖变化。方法用单细胞凝胶电泳技术,研究细胞的DNA损伤以及细胞增殖。结果在硝酸铅浓度为1.5,3,6g/l的染毒条件下,小鼠睾丸细胞核DNA拖尾为:35.297,46.205,59.023μm;胸腺细胞核DNA拖尾长为26.120,27.488,32.609μm;MTT实验结果:睾丸0.839,0.557,0.445;胸腺0.790,0.878,1.058;脾脏0.936,1.035,1.095。结论铅可引起小鼠睾丸,胸腺细胞DNA损伤,亦可致小鼠睾丸,胸腺,脾的细胞变化。从而证实铅的生殖毒性及对免疫系统的损伤作用。
【Abstract】 Objective:To study the effects of Lead on mice’s DNA damage and cell proliferation changes.Methods:Using single cell gel electrophoresis to test the cell DNA damage and cell proliferation.Results:The concentrations of lead nitrate were 1.5,3,6 g/ L of exposure conditions,the mice testis nuclear DNA tailing were:35.297,46.205,59.023μm;thymus nuclear DNA tail length were 26.120,27.488,32.609μm;MTT testing results:Testicular 0.839,0.557, 0.445;thymus 0.790,0.878,1.058;spleen,0.936,1.035,1.095.Conclusion:Lead can cause DNA damage of mice testis and thymus cell,and also can induced the cell changes of mouse testis, thymus,and spleen.Thus it was confirmed the reproductive toxicity of lead and damage of immune system.
