大鼠脊髓慢性压迫性损伤动物模型的建立

【摘要】 目的建立一种大鼠脊髓慢性压迫性损伤模型。方法应用平头不锈钢螺钉从大鼠第4颈椎(C4)椎体前侧钻入压迫脊髓腹侧,螺钉侵占脊髓深度分别为0.4、0.8、1.3mm,并规定为轻、中、重压迫。术后保留螺钉30天,并用联合行为评分(CBS)、神经电生理、大体观察、光镜、电镜检查进行综合评判。结果模型动物脊髓慢性压迫随着螺钉轻、中、重压迫程度的不同,其术后行为学、运动诱发电位(MEP)及组织学观察符合脊髓慢性压迫症病理改变的特点。结论建立了大鼠脊髓慢性压迫性损伤模型,此模型制作简便,可重复性强,脊髓损伤能表现出不同的压迫深度,为进一步研究脊髓慢性压迫损伤病理机制,研究有效治疗方药奠定了基础。

【Abstract】 Objective To establish an animal mode of chronic compressed spinal cord injury in the rat.Methods Metal screw without tip was used to dill into spinal cord through the anterior canine C4 vertebral body,then cord was chronic compressed by the screw.The depths of screw drilled into the spinal cord was 0.4,0.8、1.3mm and regarded them as mild,moderate and severe compression injury respectively.The screws were kept inside for one month after operatioa Comprehensive evaluation was carried out on CBS,elctrophysiologicl changes and observation by light microscope and electron microscope.Results The degrees of spinal cord injury varied with different depth of screws drilled into spinal cord.Postoperative behavior, Motor Evoked Potential(MEP) examination and histology changes are coindence of the character of chronic spinal cord compression myelopathy.Conclusion Techniques for creating this model is simple and can he highly replicable,and injury degrees of spinal cord can indicate the compression depth It provided an avaluable basis for the farther studies on pathological mechanism of chronic spinal cord compressed injury and effective therapies.
