沿皮浅刺内关、公孙穴对脑缺血大鼠细胞凋亡调控基因Bcl-2、Bax及血浆NOS、ET-1的影响

【摘要】 目的:研究浅刺针法对脑缺血大鼠细胞凋亡调控基因Bcl-2、Bax及血浆NOS和ET-1含量的影响的影响。方法:将120只Wistar大鼠随机分为浅刺针法组(Ⅰ组)、常规针刺组(Ⅱ组)、模型对照组(Ⅲ组)、正常对照组(Ⅳ组)。前三组采用血栓栓塞法建立脑梗塞大鼠模型。Ⅳ组不加任何处理因素,但在同一时间进行捆绑固定。每组再随机分为2、7、14天三个时间点,每个时间点各10只大鼠。通过神经体征评分、脑组织Bcl-2和Bax蛋白、血浆NOS和ET-1等几个指标,观察经过治疗后的Ⅰ、Ⅱ组大鼠和相应时间未经治疗的Ⅲ、Ⅳ组大鼠的大脑细胞凋亡情况。结果:三个时间点的浅刺针法组大鼠脑组织Bax蛋白表达明显低于模型组和常规组针刺组,而Bcl-2则明显高于模型组和常规组针刺组(P<0.01),说明浅刺针法可有效改善脑梗塞大鼠的神经缺损体征。浅刺针法组及常规针刺组的3个时间点的ET-1及NOS的表达程度均较模型对照组低(P<0.01);而浅刺针法组与常规针刺组比较则3个时间点的ET-1及NOS的表达程度均较低(P<0.01)。结论:浅刺内关、公孙穴位通过下调Bax蛋白表达和上调Bcl-2蛋白的表达,降低升高的NOS、ET-1水平,从而抑制细胞凋亡,发挥神经保护作用。并在分子水平证实经络在体表有其物质基础,浅刺可取得有效的刺激作用,使浅部组织的经络特异性能充分显示出来。

【Abstract】 Objective To study shallow Spike Act of rat brain after cerebral infarction is chemic penumbra of Bcl-2,Bax effect.Methods 120 Wistar rats were randomly divided into shallow Spike Act (Ⅰ),conventional acupuncture group(Ⅱ),the model group(Ⅲ),and normal control group(Ⅳ). Thromboembolism three groups using the rat model of the establishment of cerebral infarction.Ⅳgroup without any treatment,but at the same time bundled fixed.Then each group were randomly divided into three days after 2,7,14 point in time,each of the five time points in rats.Neural signs from the score,using immunohistochemical methods of brain tissue Fas,Bcl-2 protein expression and ET-1 and NOS expression changes observed after 2,7,14 days after treatmentⅠ,Ⅱrats and the corresponding time untreatedⅢ,Ⅳrat brain cell apoptosis.Results Three time points shallow Spike Act rat brain ischemic penumbra fas protein expression was significantly lower than the model group and the conventional group acupuncture group,and bcl-2 was significantly higher than that model and the conventional group needle thorn group(P<0.01), indicating that shallow Spike method can be effectively improved cerebral infarction in rats nerve defect signs. The remaining three groups,shallow Spike Law Unit and the routine acupuncture group 3 time points score of neurological signs,neuronal apoptosis count,ET-1 and NOS expression of the degree of model control group were low(P<0.01).And shallow barbed needle acupuncture group compared with the conventional three time points were neurological signs score,neuronal apoptosis count,ET-1 and the extent of NOS expression was lower(P<0.01).ConcIusion Bax,Bcl-2 in rat brain ischemic cerebral infarction cell apoptosis plays an important role,thorn shallow ditch,which customs very neiguan,gongsun point reduction through increased and expression of Bax protein Bcl-2 protein expression,inhibition of apoptosis,a neuroprotective effect.And Meridian confirmed at the molecular level in the body has its material foundation,shallow thorns can be an effective stimulus to shallow Meridian specific organizations can fully displayed.
