【摘要】 目的探讨色氨酸羟化酶2(TPH2)基因启动子区的-8396G/C(rs4131347)、第7外显子区的rs7305115 A/G单核苷酸多态性与抑郁症及自杀企图的关系。方法提取105例有自杀企图的抑郁症患者、102例无自杀企图的抑郁症患者和100例健康对照者的基因组DNA,应用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术,检测多态性。结果抑郁症患者和健康对照者TPH2rs4131347、rs7305115基因型和等位基因频率差异无统计学意义(P>0.05);有自杀企图的抑郁症患者和无自杀企图的抑郁症患者TPH2 rs4131347基因型和等位基因频率差异无统计学意义(P>0.05);rs73051 15位点基因型及等位基因分布频率在抑郁症有、无自杀企图组之间的差别有统计学意义(P<0.05)。抑郁症有自杀企图组内的个体携带TPH2 rs7305115基因型AA的频率及等位基因A的频率均较低,差异有统计学意义(P<0.01)。结论 TPH2基因rs4131347、rs7305115单核苷酸多态性与抑郁症无明显关联。TPH2基因rs73051 15位点A/A基因型和A等位基因可能与抑郁症的自杀企图有关联,其可能是抑郁症的自杀企图的保护因素。更多还原

【Abstract】 Objective To explore the association between polymorphisms of Tryptophan hydroxylase2 gene promoter region or seventh exon region,major depressive disorder and suicidal plan.Methods The allele and genotype distribution of TPH2 gene SNP rs4131347 rs7305115 were detected in 105 depression patients with suicidal plan,102 depression patients with no suicidal plan and 100 healthy control subjects by using polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP),so as to determine the association between TPH2 and depression or suicidal plan.Results The gene polymorphism at rs4131347 and rs7305115 were indefinite.No significant difference in the allele and genotype distribution at TPH2 rs4131347、 rs7305115 was found between the depressive patients and healthy control group(P > 0.05).No significant difference in the allele and genotype distribution at TPH2 rs4131347 was found between the depressive patients with suicidal plan and the depressive patients with no suicidal plan(P>0.05).The significant difference in the allele and genotype distribution at TPH2 rs7305115 was found between the depressive patients with suicidal plan and the depressive patients with no suicidal plan(P < 0.05).The A allele and AA genotype distribution in the depressive patients with suicidal plan is low.Conclusion No correlation was found between TPH2 gene SNP rs4131347、 rs7305115 and major depressive disorder.The findings support that the A allele and A A genotype of rs7305115 A/G at TPH2 gene seventh exon region in Han nationality is associated with suicidal plan of depressive patients and may be protection factor of suicidal plan in depressive patients.更多还原
