不同种植年限铁皮石斛多糖、甘露糖含量的测定及其它化学成分比较研究

【摘要】 目的:比较不同种植年限铁皮石斛的多糖及甘露糖含量,并对其正丁醇部位及乙醚部位化学成分进行比较研究,为铁皮石斛种植年限的确定提供参考。方法:采用苯酚-硫酸法测定多糖含量,柱前衍生化HPLC法测定甘露糖的含量,RP-HPLC结合二极管阵列检测器(DAD)法分析铁皮石斛正丁醇部位(360nm)及乙醚部位(280nm)分别随不同种植年限的变化情况。结果:铁皮石斛多糖含量随种植年限不同而变化,2年生>3年生>4年生,2~4年生铁皮石斛随种植年限增加,多糖含量减少。甘露糖含量随种植年限的变化也呈现与多糖相同的变化趋势。360nm下铁皮石斛正丁醇部位中黄酮类成分随种植年限变化有一定的变化,2年生中黄酮类化合物相对极性较小,且较集中。3年生中极性分布较广,且含量相对较大。4年生主要分布在极性大的黄酮类化合物,含量相对较低。280nm下铁皮石斛乙醚部位随着种植年限增加,色谱图峰强度增加明显。结论:从多糖及甘露糖含量考虑,以2年生铁皮石斛含量最高,宜以两年生为用;而结合酚类及黄酮类成分却与此有所背离,具体有待进一步深入研究。
【Abstract】 OBJECTIVE:To compare the polysaccharideand mannose content in different cultivating years of D.officinale Kimura etMigo and chemical compositionof n-butanol and ether portion.METHODS:The content of polysaccharides and mannosefrom Dendrobiumwere determined by phenol-sulfuric acid method and pre-column derivatization HPLC.Chemical compositionanalysis of n-butanol and ether portion for different cultivating years of D.officinale KimuraetMigo was carried on rp-HPLC equipped with DAD at 360 nm and 580 nm.RESULTS:The results showed that the content of polysaccharides and mannose was decreasing as growth years increased,among which the content of 2 years was the highest.The peak intensity of flavonoids at 360 nm from n-butanol was variable with growth years.While the total peak intensity of ether portion at 280 nm keeps increasing along withgrowth years.CONCLUSIONS use the polysaccharide content and mannose content as the mainreference index of quality,it’ssuitable to harvest.the D.officinale Kimura et Migo of two years old.Whereas,considering the flavonoids and total phenols,two years is possibly not a best choice,which deserves further investigation.
