【摘要】 目的:研究补肾益气方药对自然衰老大鼠CD4~+T细胞NF kappa B核转位与IL2基因转录的影响。方法:以自然衰老大鼠为模型,分组:青年对照组;老年对照组;老年补肾益气组。流式细胞仪分选各实验组大鼠CD4~+T细胞,以anti-CD3联合anti-CD28进行刺激。Q Realtime RT-PCR法检测IL-2 mRNA的表达;ELISA法检测血清IL2蛋白浓度;激光共聚焦检测转录因子NF-κB细胞内定位;Western blotting法检测NF-κB在胞浆及核内的表达。结果:与青年对照组比较,anti-CD3联合anti-CD28刺激后老年对照组CD4~+T细胞IL-2 mRNA表达及血清浓度均显著降低(P<0.01);刺激0.5h时,胞浆NF-κB表达明显增高,核内NF-κB表达明显降低。与老年对照组比较,老年补肾益气组CD4~+T细胞IL-2 mRNA表达及血清浓度均显著升高(P<0.01);刺激0.5h时,胞浆NFκB表达明显降低,核内NFκB表达明显升高(P<0.05)。结论:老年大鼠CD4~+T细胞对外界刺激(anti-CD3联合anti-CD28)的敏感性下降,使细胞内NF-κB核转位能力减弱。补肾益气方能通过活化衰老CD4~+T细胞NF-κB的核转位,增强NF-κB与IL-2基因的结合,提高IL-2表达,延缓衰老细胞免疫功能衰退。更多还原

【Abstract】 Objective:To investigate whether the Bushenyiqi Formula(BSYQ) can elevate the IL-2 expression through regulating the NF kappa B activity in the aged rat CD4~+T Cells.Methods:We analyzed the IL-2 expression in CD4~+T Cells from different aged rats(young group:around 6 months(n=6),aged group:around 23 months(n=6),aged BSYQ group:around 23 months(n=6).), the translocation activity of NF-κB and its protein level in the cytoplasm/ nucleus at different time points,using confocal microscopy and western blotting.Results:Our results showed,aged cells synthesis lower IL-2 than the young CD4~+T Cells(P<0.01).Less NF-κB was translocated into nucleus markedly at stimulated for 0.5h compared with that in the young group.Compared with the aged group,after treated with BSYQ formula,the translocation ability of NF-κB elevated,thereby up-regulated the expression of IL-2更多还原
