IL-17和IL-23在肥胖哮喘小鼠气道炎症中的作用及其与氧化应激的关系

【摘要】 目的探讨Th-17相关因子——IL-17和IL-23在肥胖哮喘小鼠气道炎症中的作用及其与氧化应激反应的关系。方法45只雌性C57/6J小鼠随机分为哮喘组、肥胖哮喘组(肥哮组)和对照组各15只。卵白蛋白激发致敏和高脂饮食制作肥胖哮喘模型,计数支气管肿泡灌洗液(BALF)中细胞总数并分类,酶联免疫吸附法法测定肺组织匀浆中白细胞介素(1L)-17、IL-23和8-异前列腺素2α(8-iso-PGF2α)水平。结果哮喘组小鼠BALF中白细胞总数和嗜酸粒细胞比例均较对照组明显升高(P<0.05)。肥哮组肺组织匀浆的IL-17、IL-23和8-iso-PGF2α水平分别为(65.83±5.69)pg/ml、(141.52±14.62)pg/ml和(61.33±7.75)pg/ml均高于哮喘组和对照组(P<0.05)。Pearson相关分析表明,IL-17与8-iso-PGF2α水平存在相关性(r=0.758,P<0.01)。结论肥胖哮喘小鼠体内IL-17和IL-23水平升高,参与了哮喘气道炎症和氧化应激过程,抑制Th17偏移可望成为肥胖哮喘治疗的有效措施。

【Abstract】 Objective To evaluate the effect of T help cell 17(Thl7) on the airway inflammation and oxidative stress of obese asthma mice with high-fat diets.Methods Forty-fives female C57BL/6J mice were randomly divided into 3 groups,asthma group,obese asthma group and control group.Asthma group were sensitized and challenged by ovalbum(OVA) and normal diets,obese asthma group were sensitized,challenged as asthma group and fed with high-fat diets,the control group were sensitized,challenged with normal saline and fed with normal diets.The cells in BALF were count and classified after staining,IL-17,IL-23 and 8-iso-prostaglandinF2a(8-iso-PGF2a) in lung tissue were detected by enzyme linked immunosorbent assay.All the parameters were compared between 3 groups.Results In comparison with control group,the leukocytes and EOS count in BALF were increased as well as 8-iso-PGF2 a in lung tissue elevated in asthma group.1L-17,IL-23 and 8-iso-PGF2 a in lung tissue of the obese asthma mice were(65.83±5.69)pg/ml,(141.52±14.62)pg/ml and(61.33±7.75) pg/ml separately,all of which were increased than those in asthma group(P<0.05).Pearson correlation analysis showed that IL-17 were in positive correlation with 8-iso-PGF2α(r= 0.75 8,P<0.01).Conclusion Th17 related cytokines were involved in the airway inflammation and oxidative stress of obese asthma mice with high-fat diets.
