电针对慢性时差模型金黄地鼠下丘脑CRH基因表达影响研究

【摘要】 目的:探讨电针对慢性时差模型金黄地鼠神经内分泌应激轴的调节作用。方法:运用超前性光暗周期转移方法,复制慢性时差金黄地鼠模型。造模结束后,恢复正常光照制,选用ZT4(zeitgebertime,12:00)电针"百会"、"长强",电压1-3v,频率15-25Hz,时间15min,针刺3天,1次/天,运用荧光实时定量RT-PCR检测下丘脑CRH基因表达,采用酶联免疫法检测血清CORT含量。结果:慢性时差造模后,以及慢性时差造模自然恢复3天后,动物下丘脑CRH基因表达较正常有所降低,血清CORT含量较正常显著增高;针刺对下丘脑CRH基因表达、CORT含量均有恢复趋势。结论:电针在一定程度上有调节慢性时差模型动物HPA轴功能的作用。
【Abstract】 Objective:The aim of this study was to investigate the effect of Electro-acupuncture(EA) on neuroendocrine HPA axis in Syrian hamster model of chronic Jet Lag.Methods:Model of chronic jet lag was established by appling the methods of advances of light/dark cycle for 10 days.EA(15/25Hz in frequency, l-3v) was applied to "Baihui"(Du20) and "Changqiang"(Du1) for 15min/d and lasted for 3 days.The expression of CRHmRNA was measured by PR-PCR method.The content of CORT was detected by ELISA method.Results:(1) Compared with the control group,CRHmRNA in the hypothalamus was decreased,and CORT in serum was significant increased in the model group and the Natural recovery group.(2) Compared with the model group and the Natural recovery group,CRHmRNA in the hypothalamus and CORT in serum were displayed a recovery trend by EA.Conclusion:The effect of EA may be correlated with regulating HPA axis function.
