【摘要】 <正>目的 :研究芍药苷对人T淋巴细胞增殖、凋亡及产生Th1/Th2型细胞因子IFN-γ/IL-4的影响。方法:分离健康人外周血T淋巴细胞,抗人CD3 Ab+抗人CD28 Ab共刺激T淋巴细胞活化,SRB法检测芍药苷对T淋巴细胞增殖的抑制,流式细胞术检测芍药苷诱导T淋巴细胞凋亡,ELISA法分析芍药苷对T淋巴细胞上清中细胞因子IFN-γ/IL-4的影响。结果:芍药苷可呈浓度依赖性抑制T淋巴细胞增殖(P<0.01);芍药苷可诱导T

【Abstract】 Objective:To study the effects of paeoniflorin on human peripheral blood T lymphocytes proliferation,apoptosis and the Th1 and Th2 type cytokine IFN -γ/IL - 4 level in supernatant.Methods:Human peripheral blood T lymphocytes were isolated and activated by anti - CD3 mAb plus anti - CD28 mAb.The inhibition of paeoniflorin on T lymphocytes proliferation was measured by Sulforhodamine B(SRB) method.Apoptosis was analysed by flow cytometry and the cytokine IFN -γ/IL -4 level in supernatant were measured by ELISA.Results;Different doses of paeoniflorin inhibited proliferation and induced apoptosis of these cells after costimulated by anti - CD3 mAb and anti - C28 mAb in dose - dependent manner(P<0.01).Paeoniflorin lowered IFN -γlevel in T lymphocytes supernatant(P<0.01 ), but had no effects on IL -4 level(P>0.05).Conclusion;Paeoniflorin inhibited proliferation and induced apoptosis ol human peripheral blood T lymphocytes after costimulated by anti - CD3 mAb plus anti - C28 riiAb in dose - dependent manner.Paeoniflorin decreased IFN -γlevel in T lymphocytes supernatant in dose - dependent manner.The anti - inflammation mechanism may be related to apoptosis of T lymphocytes and decrease in IFN -γlevel.
