喘可治注射液诱导哮喘小鼠Foxp3~+调节性T细胞的免疫机制探讨

【摘要】 目的探讨喘可治注射液诱导Foxp3~+调节性T细胞(Foxp3~+Tr)在防治变应性哮喘的免疫调节机制。方法将Balb/C小鼠分为对照组、哮喘组、喘可治(CKZ)组三组,采取以OVA经腹腔注射致敏加滴鼻激发复制变应性哮喘模型,CKZ组给予喘可治注射液腹腔注射治疗,对照组与哮喘组给予生理盐水腹腔注射。分别观察各组小鼠的肺部病理改变,流式细胞术检测CD4、CD25、Foxp3的百分比,RT-PCR检测Foxp3、TGF-β、IL-10mRNA表达情况。结果 1.哮喘组较对照组表现出明显炎症反应,CKZ组较哮喘组气道炎症减轻;2.哮喘组下调脾脏CD4~+T、CD4~+CD25~+Tr、Foxp3~+CD4~+CD25~+Tr比例;CKZ组上调CD4~+T、CD4~+CD25~+Tr、Foxp3~+CD4~+CD25~+Tr比例;3.哮喘组上调肺组织Foxp3、IL-10、TGF-βmRNA;CKZ组下调Foxp3、IL-10、TGF-βmRNA。结论哮喘小鼠表现出免疫异常的状态,喘可治注射液能够通过诱导Foxp3+调节性T细胞的生成,调整机体紊乱的免疫状态,使机体的免疫系统恢复平衡状态,从而发挥抑制炎症的免疫治疗作用。
【Abstract】 Objective To investigate the immune regulation mechanism of Chuankezhi(CKZ) on allergic asthma treatment.Methods Mice were sensitized and challenged with ovalbumin(OVA) to establish allergic asthma - like model.To determine the contribution of CKZ on airway allergic inflammation and immune regulation,we examined the lung histopathology,cytometric analysis of spleen CD4~+ CD25~+ Foxp3~+ cells by FCM,and expression of Foxp3,TGF -beta and IL - 10 mRNA in the lung tissue by RT - PCR.Results CKZ treatment suppressed murine airway eosinophilia, peripheral vascular congestion and alveolar compensatory expansion,up - regulated spleen CD4~+ T,CD4~+ CD25~+ Treg cells and Foxp3~+ CD4~+ CD25~+ Treg cells,down - regulated lung tissue foxp3,IL - 10,TGF - B mRNA expression, as compared with asthma group.Conclusions Asthmatic mice showed a abnormal immune status.CKZ can restore balance to the abnormal immune status by induing Foxp3 + regulatory T cells so as to play the role of immune therapy to suppress inflammation.
