针刺对AD模型小鼠海马脑红蛋白表达的影响

【摘要】 目的:探讨"补肾活血针刺法"对阿尔茨海默病模型小鼠SAMP8(SenescenceAccelerated Mouse P8,SAMP8)学习记忆能力改善作用的物质基础和潜在机制。方法:将5月龄雄性SAMP8小鼠20只随机分为针刺治疗组(n=10)、模型对照组(n=10),另设5月龄雄性SAMR1小鼠(n=10)作为正常对照组。针刺治疗组予以"补肾活血针刺法"干预;模型对照组以及正常对照组未予特殊处理;干预4周后,取各组小鼠海马组织,采用免疫组化法检测脑红蛋白的表达情况。结果:模型对照组SAMP8小鼠海马组织Ngb的表达水平较正常对照组SAMR1小鼠减弱(p<0.001);与模型对照组相比,针刺治疗组小鼠海马组织Ngb的表达水平明显升高(p<0.001)。结论:针刺干预能够明显增强SAMP8小鼠海马组织Ngb的表达,这可能是其改善SAMP8小鼠学习记忆能力的潜在分子机制之一。
【Abstract】 Objective:To investigate the material base and underlying mechanism of the therapeutic effect on senescence-accelerated mice P8(SAMP8) which is considered as an generally accepted mouse model of Alzheimer’s disease by acupuncture.Methods:20 five-month-old male SAMP8 were randomly divided into two groups:acupuncture treatment group(n=10) and model control group(n =10).Another 10 five-month-old male SAMR1 mice were prepared as normal control group(n=10).Mice in acupuncture treatment group were treated by acupuncture with reinforcing kidey and activating blood method for 4 weeks and Baihui(GV20),Shenshu(BL23),Xuehai(SP10) and Geshu(BL17) were selected,mice in model control group and normal control group were raised without special treatment.After 4 weeks,the expression of neuroglobin in hippocampus tissue were examined by immunohistochemistry.Result:The expression levels of Ngb significantly decreased in SAMP8 mice of model control group compared with SAMR1 mice of normal control group(p<0.001);Compared with mice of model control group,the expression level of Ngb in acupuncture treatment group was significantly increased(p<0.001). Conclusion:Acupuncture could increase the expression of Ngb in hippocampus tissue of SAMP8,which may be one of the underlying molecular mechanisms in improving learning and memory abilities of SAMP8 by acupuncture.
