【摘要】 目的:研究经前平颗粒对经前期综合征(PMS)肝气逆证模型大鼠不同脑区(下丘脑、顶区皮质)孕酮受体(PR)表达的影响。方法:以情志刺激多因素法复制PMS肝气逆证大鼠模型,以经前平颗粒进行干预,采用免疫荧光标记结合激光流式细胞仪、激光共聚焦显微镜检测大鼠下丘脑和顶区皮质中PR的蛋白表达量及定位分布模式。结果:模型组大鼠下丘脑和顶区皮质中PR蛋白表达水平与正常组相比均显著降低(P<0.05),经前平颗粒干预后该受体表达水平较模型组显著上调。结论:PMS肝气逆证发病与下丘脑和顶区皮质中PR含量密切相关,经前平颗粒显著上调两部位PR蛋白表达可能是其治疗PMS的机制之一。更多还原

【Abstract】 Objective:To study the effects of Jingqianping granule on the expression levels of progesterone receptor(PR) in different brain regions(hypothalamus and parietal cortex)of premenstrual syndrome(PMS) model rats with liver-qi invasion.Methods:The PMS model with liver-qi invasion was induced in rats by multiple emotional stimulations,and then was administered with Jingqianping granule. The expression levels of progesterone receptor and its’ Location and distribution pattern in hypothalamus and parietal cortex were detected by immunofluorescence technique coupled with flow cytometry and laser scanning confocal microscope.Results:The expression levels in hypothalamus and parietal cortex of progesterone receptor were significantly lower in PMS model rats than in control rats(P<0.05), Jingqianping granule markedly increased the reduced expression levels.Conclusion:The content of progesterone receptor in hypothalamus and parietal cortex is closely related to the pathogenesis of PMS with liver-qi invasion,maybe the significant up-regulation of progesterone receptor expression level in these two regions is one of the action mechanisms of Jingqianping granule.更多还原
