芪麝丸对直立大鼠腰椎骨形成的作用观察

【摘要】 目的观察芪麝丸对直立体位大鼠腰椎骨形成的作用,探讨可能的作用机制。方法90只SD大鼠随机分为三大组,正常组不进行处理,普通饲养笼喂养;模型组和芪麝丸组建立长期直立体位大鼠模型。每大组再随机分为5月,7月和9月组,用药组分别在术后4,6,8个月开始灌胃,连续灌胃一月后取材,L5椎体藏红染色,Ⅰ型胶原(typeⅠcollegan,ColⅠ)免疫组织化学染色,余下腰椎椎体应用实时荧光定量聚合酶链反应(real time reversetranscription polymerass chain reaction,real time RT-PCR)技术检测Collα2、转化生长因子beta1(transform growth factor beta1,TGF-β1)和一种核转录因子(core bindingfactor a1pha1,Runx2)基因的表达。结果藏红染色显示,与正常组比较,三个时间点模型组椎体骨小梁均增加,用药组椎体骨小梁与模型组类似。ColⅠ免疫组织化学染色显示9月用药组骨小梁数目和厚度均较模型组增加,阳性染色范围增加,较正常组深染。Real time RT-PCR示,与模型组比,7月用药组Collα2、TGF-β1、Runx2基因表达都显著上调,5月用药组Collα2、TGF-β1基因表达显著下调(P<0.05,P<0.01),9月用药组Collα2基因表达显著下调(P<0.05),而rGF-β1和Runx2基因表达均显著上调(P<0.05)。结论芪麝丸对直立增加的椎体成骨有增强作用,以中后期(7月、9月)效应显著。

【Abstract】 Objective To observe the effect of Qi She Wan on upright posture rats’ lumbar vertebral bone formation and investigate its potential mechanism.Methods Ninety SD rats were randomly divided into three groups.The rats had no treatment in control group,and were raised in normal cage.The rats were induced to upright posture in model and Qi She Wan groups.Every group were randomly divided into three secondary groups:5 month,7 month and 9 month group.Rats in Qi She Wan groups were given intragastric administration 4,6, 8 months after surgery.Ten rats were sacrificed at 5 month,7 month and 9 month after surgery and all lumbar vertebras were harvested for detection.Safranine O/fast green staining and ColⅠimmunohistostaining were performed on L5 vertebrae.Col1α2、TGF-β1 and Runx2 mRNA expression in other vertebras were detected by Real time RT-PCR.Result Safranine O/fast green staining shows in three model groups,bone trabecular density increased and similar changes in Qi She Wan groups.ColⅠimmunohistostaining shows bone trabeculaes increased and thickened,positive staining extended in 9 month Qi She Wan group compared with those in model group.ColⅠpositive staining was also deeper in 9 month Qi She Wan group than that in control group.Real time RT-PCR shows Col1α2、TGF-β1 and Runx2 mRNA expression was obviously up-regulated in 7 month Qi She Wan group compared with that in 7 month model group.Col1α2 and TGF-β1 mRNA expression was obviously down-regulated in 5 month Qi She Wan group compared with that in 5 month model group(P<0.05,P<0.01).Col1α2 mRNA expression was obviously down-regulated in 9 month group Qi She Wan group compared with that in 9 month model group(P<0.05).Conclusion Upright posture may increase rats’ vertebral bone formation and Qi She Wan may strengthen the effect,especially at midanaphase(7 month and 9 month group).
