方向性冰晶低温试验台研究

【摘要】 在细胞的冻结保存过程中,冰晶损伤是导致细胞死亡的主要因素之一,如何减小冰晶的损伤是细胞成功保存的关键。本文研制的方向性冰晶实验台可以改变冰晶的形态,为研究细胞的冰晶损伤提供了工具。用方向性冰晶实验台冻结不同浓度的MeSO2保护剂,并用显微镜观察在不同降温速率下冰晶生长情况,结果表明,影响冰晶结构形态的主要因素是冷却速率和浓度,随着冷却速率的增大,冰晶变的细而尖;随着浓度的增大,冰晶的尺寸也变大。
【Abstract】 Ice damage is one of the factors that causes cell death in the cryopreservation process. This paper developed a directional ice formation cryostage to study the ice damage to cells. Various MeSO2 solutions were frozen on the cryostage. The ice was inspected by a microscope and recorded by a digital camera. It was shown that the size and structure of ice were affected by cooling rates and concentration of the solution. The ice became fine and sharp with the increment f cooling rates, and the size became larger with the increment of concentration.
