【摘要】 目的:研究注射用双黄连对Hartley豚鼠与BN大鼠诱导的主动全身过敏反应(Active Systemic Anaphylaxis,ASA)的差异,为完善中药注射剂过敏性评价提供实验依据。方法:取Hartley豚鼠或BN大鼠随机分为注射用双黄连、卵蛋白(OVA)、阴性对照组。分别给予皮下注射双黄连豚鼠、大鼠致敏液800、750mg/kg,卵蛋白豚鼠、大鼠致敏液5mg/只,进行Hartley豚鼠与BN大鼠ASA试验,检测激发后30min动物的全身过敏反应症状、HE染色观察肺组织病理改变、放免法检测BN大鼠在致敏前、致敏第18天以及激发后血清总IgE水平的变化。结果:对Hartley豚鼠或BN大鼠,卵蛋白组动物呈现强阳性过敏反应症状,注射用双黄连组动物呈现弱阳性过敏反应症状。肺组织病理切片显示:卵蛋白、注射用双黄连组动物肺组织血管周围均有明显淋巴细胞浸润等炎症病理表现。与阴性对照组相比,卵蛋白、注射用双黄连组大鼠给药致总IgE水平显著升高(P<0.01)。与致敏前比较,卵蛋白、注射用双黄连组大鼠总IgE水平显著升高(P<0.01)。结论:增加不同品种实验动物及相关检测指标,能更全面评价注射用双黄连潜在致敏性。更多还原

【Abstract】 Objective:To set up multiple criterions of active systemic anaphylaxis(ASA) induced by Shuanghuanglian injection.Methods:Hartley guinea pigs or BN rats were randomly divided into OVA group,Shuanghuanglian injection group and negative control group.Animals were injected with different drugs or saline at 0.5 ml each on first,third,fifth day and repeated on nineteenth day.The usual systemic symptoms of ASA test were observed within 30min after the last injection. The lung tissue pathological examination was performed with HE staining.Serum IgE levels were determined before,18 days and 19 days after first injection using radioimmunoassay.Results: Hartley guinea pigs or BN rats in OVA group and Shuanghuanglian injection group showed positive systemic allergy symptoms.However,pathological changes of the lung tissue with perivascular lymphocyte infiltration were observed and higher levels of serum IgE were determined both in OVA group and Shuanghuanglian group of guinea pigs or BN rats.Conclusion:The multiple criterions of evaluation with different animal species and increased observation targets for the potential ASA detection induced by Shuanghuanglian injection are more comprehensive and acceptable.更多还原
