鹿茸草中苯乙醇苷类化合物的抗菌抗炎作用

【摘要】 目的:研究中药鹿茸草中苯乙醇苷类化合物(TPG)的抗菌抗炎活性。方法:本研究从鹿茸草中提取分离鉴定了4种苯乙醇苷类化合物。首先考察TPG的抑菌作用,确定TPG的最小抑菌浓度(MIC)和最低杀菌浓度(MBC);利用小鼠全身感染模型,观察TPG对感染小鼠存活期的影响;采用小鼠鼻腔吸入绿脓杆菌致肺炎模型,考察TPG对肺炎小鼠肺匀浆液培养后菌落生长情况及组织病理变化;分别采用二甲苯致小鼠耳肿胀及棉球肉芽肿模型考察TPG的抗炎作用。结果:体外抗菌实验中TPG对各菌株都有明显的抗菌作用,其MIC值为16 0.0625-0.5000 mg/ml,绿脓杆菌、白色念珠菌和金黄色葡萄球菌MBC值分别为2、8、16 mg/ml;在绿脓杆菌和金黄色葡萄球菌致小鼠全身感染实验中,TPG(60,120,180 mg/kg)给药组的存活率分别为37.5%、50%和87.5%,20%、40%和80%;在绿脓杆菌致肺炎模型中,TPG(180mg/kg)能显著减少感染小鼠肺部细菌数,与模型组相比存在显著性差异(P<0.05),并减轻了肺组织的病理改变,减少了炎性细胞浸润。在二甲苯致小鼠耳肿胀实验中,TPG(180mg/ml)与模型组相比存在显著性差异(P<0.05):小鼠肉芽肿实验结果中,TPG(120mg/ml、180mg/ml)与模型组相比存在显著性差异(P<0.01)。结论:TPG具有显著的体内外抑菌抗炎作用。

【Abstract】 Aims:To investigate the antibacterial activity,anti-inflammatory of total phenylethanoid glycosides(TPG) from Monochasma sauatieri Franch.Methods:The anti-bacterial effects of TPG were evaluated by determining the minimum inhibitory concentration(MIC) and minimal bactericidal concentration(MBC) in vitro.The antibacterial activities of TPG in vivo were examined by using a Pseudomonas aeruginosa or Staphylococcus aureus induced sepsis model to calculate the mortality of mice and Pseudomonas aeruginosa induced pneumonia model to count the colonies and examine the histopathological change.Additional,xylene-induced ear edema and cotton pellet implantation in mice were established to demonstrate the anti-inflammatory activities of TPG.Results:TPG showed in vitro antimicrobial activity against each of the tested strains with MIC values rang from 0.0625 to 0.5000 mg/mL,and the MBC of TPG was 2 mg/ml {Pseudomonas aeruginosa) and 16 mg/ml {Staphylococcus aureus) respectively.In vivo,TPG(60, 120,180 mg/kg)-treated groups showed significantly high survival rates of 37.5,50 and 87.5% respectively in Pseudomonas aeruginosa-induced sepsis,meanwhile,in Staphylococcus aureus-induced sepsis survival rate of animal was 20,40 and 80%respectively.Besides,in the Pseudomonas aeruginosa infection pneumonia,the results indicated that TPG(180 mg/kg) had bacterial clearance activity comparedwith control group(P<0.05) and revealed significantly milder inflammatory cells infiltration and proliferation in comparison to control group.Moreover, in the anti-inflammatory experiments,TPG(180 mg/kg) significantly inhibited ear edema,when compared with the control group(P<0.05) and TPG(120,180 mg/kg) significantly inhibited granulomatous tissue formation in cotton pellet at a dose-dependent manner,with inhibition rate of 24.6%and 29.7%(P<0.01),respectively.Conclusion:TPG might have the potential abilities to be promising natural medicine for chronic Pseudomonas aeruginosa and Staphylococcus aureus lung infections.In addition,it also provided evidence to rationalize the traditional use of Monochasma savatieri Franch for inflammation.
