大鼠前额叶皮层神经元的急性分离及应用

【摘要】 目的建立一种稳定的大鼠前额叶皮层(Prefrontal Cortex,PFC)神经元的急性分离方法,用于中枢神经元离子通道特性的研究。方法低温人工脑脊液灌流后,结合酶消化及机械吹打,急性分离6～40天鼠龄大鼠的前额叶皮层神经元。采用全细胞膜片钳技术记录电压门控钠电流和配体门控甘氨酸激活电流。
【Abstract】 Objective To establish a reliable method for isolation of single neuron from the prefrontal cortex(PFC) of rodent for patch clamp electrophysiological recordings. Methods After preparing the brain slices from young rats,the neurons from prefrontal cortex were acutely isolated using enzyme digestion and mechanical trituration.The neurons were then used to record both the voltage-gated currents and ligand-gated currents using the whole cell patch clamp technique.
