鞘内注射p-MPPF对异氟烷镇痛作用的影响

【摘要】 目的探讨异氟烷(isoflurane,Iso)镇痛作用是否与激动小鼠脊髓5-HT1A受体有关。方法使用甩尾法、热板法、扭体法。3种方法共筛选出240只小鼠,每种方法80只,均分为以下8小组:生理盐水组(NS组),异氟烷组(Iso组),单用不同剂量5-HT1A体拮抗剂p-MPPF组(MPPF6组、MPPF3组、MPPF1.5组),合用药组(Iso+MPPF6组、Iso+MPPF3组、Iso+MPPF1.5。甩尾法以甩尾潜伏期,热板法以舔后足潜伏期,扭体法以15分钟内扭体次数为指标,观察鞘内注射(intrathecal injection,i.t)p-MPPF对Iso镇痛作用的影响。结果单独i.t 6μg、3μg、1.5μgp-MPPF对上述观察指标无明显影响(p>0.05)。与Iso组相比,Iso+MPPF6组的甩尾潜伏期和舔后足潜伏期均缩短(p<0.01),15分钟内的扭体次数增多(p<0.05);Iso+MPPF3组甩尾潜伏期和舔后足潜伏期均缩短(p<0.01或p<0.05),扭体次数无明显变化(p>0.05);Iso+MPPF1.5组的三种观察指标均无统计学差异(p>0.05)。结论吸入麻醉药Iso镇痛作用与激动脊髓5-HT1A受体密切有关。
【Abstract】 Objective The present study was designed to investigate whether isoflurane(Iso) produces the analgesic effects by exciting the 5-HT1A receptor in the spinal cord of mice.Methods Tail-flick test, hot-plate test and acetic-acid-writhing test were used.Each test required 80 mice that meet the requirements so there was a total of 240 mice.80 mice were divided into the following eight groups:NS group,isoflurane group(Iso group),p-MPPF(5-HT1A receptor antagonist) alone in three different dose groups(MPPF6 group,MPPF3 group,MPPF1.5 group),combined treatment groups(Iso+MPPF6 group, Iso+MPPF3 group,Iso+MPPF1.5 group).The tail-flick latency(TFL) in tail-flick test,the pain threshold in hot-plate test(HPPT),writhing times in 15 minutes as indicators to observe the impacts of p-MPPF intrathecally(i.t) on the analgesic effects of Iso.Results I.t 6μg,3μg,1.5μg p-MPPF alone had no effects on the observed indicators(p>0.05);Compared with Iso group,the TFL and HPPT of Iso+MPPF6 group were shortened(p<0.01),the writhing times in 15 minutes were increased(p<0.05); The TFL and HPPT of Iso+MPPF3 group had been shorten(p<0.01 or p<0.05) but writhing times had no significant changes(p>0.05);the three kinds of indicators of Iso+MPPF1.5 had no statistical difference (p>0.05).Conclusion These findings suggeste that the analgesic effects of Iso are closely related to the 5-HT1A receptor in the spinal cord.
