瑶药“猛老虎”对免疫性肝纤维化的作用研究

【摘要】 目的观察猛老虎对大鼠免疫性肝纤维化的影响。方法采用猪血清腹腔注射建立大鼠免疫性肝纤维化模型,通过观察肝组织Ⅰ、Ⅲ型胶原、TGF-β1、Hyp、MMP-1和TIMP-1含量,血清PCⅢ、LN、HA、IFN-γ、TNF-α等指标的变化,以及各组大鼠肝组织的病理学改变,对猛老虎石油醚、乙酸乙酯、乙醇和水溶4个部位进行抗肝纤维化药效学试验筛选,以判断猛老虎具有抗肝纤维化活性作用的部位。结果猛老虎乙酸乙酯部位能减少大鼠血清PCⅢ、LN、HA和TNF-α含量,提高IFN-γ水平;减少大鼠肝组织Ⅰ、Ⅲ型胶原、TGF-β1相对表达和Hyp、TIMP-1含量,促进MMP-1生成,效果与秋水仙碱相当。肝脏病理组织观察亦证实上述结果。结论猛老虎具有抗肝纤维化活性的有效部位主要存在于乙酸乙酯部位。

【Abstract】 Objective To study the effect of"Meng Laohu"on the immunity hepatic fibrosis in rats.Methods The model rats of hepatic fibrosis were induced by intraperitoneal injection of porcine serum.To judge the part in"Meng Laohu"which has the effects on anti-hepatic fibrosis from the petroleum ether part、the ethyl acetate part、the ethylalcohol part and the water-dissolve part by pharmacodynamic screening test.To observe the content of col-Ⅰ、col-Ⅲ、TGF-β1、Hyp、MMP-1 and TIMP-1 in rats’ liver tissue、the change of PCⅢ、LN、HA、IFN-γ、TNF-αet al in rats’ blood serum and the change of histopathology in rats’ liver.Results The results showed that the ethyl acetate extract of MengLaoHu can significantly decreased the contents of PCⅢ、LN、HA and TNF-α,and increased the content of IFN-γ,can down-regulate the expression ofⅠ,Ⅲ-type collagen、TGF-β1、the content of Hyp and TIMP-1,and can obviously promote the levels MMP-1,Its effect was equivalent to that of the colchicine.This result was also confirmed by liver histopathology.Conclusions The effective part in"Meng Laohu"of anti-hepatic fibrosis mainly exist in ethyl acetate part.
