通塞脉片对大鼠实验性动脉粥样硬化模型血管内皮细胞的影响

【摘要】 目的研究通塞脉(TSM)对大鼠实验性动脉粥样硬化(AS)内皮细胞的影响,并探讨其作用机制。方法50只雄性wistar大鼠,随机分5组:空白组、模型组、辛伐他汀组、TSM高剂量组、TSM低剂量组,除空白对照组给以普通饲料喂养外,其他组均饲以高脂饲料及维生素D3,建立食饵性动脉粥样硬化模型。12周后检测外周血循环内皮细胞(CEC)、血浆血管紧张素Ⅱ(AngⅡ)、电镜观察动脉血管形态。结果模型组大鼠CEC明显增加,AngⅡ含量显著升高,TSM能够减少CEC,降低AngⅡ含量。透射电镜观察模型组大鼠主动脉内皮(EC)严重受损;TSM组大鼠主动脉EC扁平形态基本完整,内弹性膜平整厚度均匀,损伤明显减轻。结论TSM通过减少CEC数量、降低AngⅡ水平,达到保护大鼠实验性动脉粥样硬化模型大鼠内皮细胞的作用。

【Abstract】 Objective To investigate the effect of TSM tablet on atherosclerosis model’s EC of rat and explore the mechanism of it.Methods 50 wistar rats were randomly divided into five groups:control group,model group, Simvastatin group,TSM high group,TSM low group.All groups were fed with high fat diet and vitamin D3 except control group to set up model of atherosclerosis.After 12 weeks we detected circle endothelial cell,angiotensinⅡand electron microscopy morphology of arteries.Results CEC and AngⅡin model rats were significantly higher. TSM can reduce the CEC and AngⅡ.Model group rat’s artery endothelial cells were severely damaged by using electron microscopy;TSM group rat’s artery endothelial cells were basic integrity and internal elastic membrane formation were thickness.It significantly reduced the injury.Conclusion TSM by reduceing the number of CEC,the level of AngⅡwhich is used for experimental atherosclerosis’s EC in rats have protective effect.
