龙胆对大鼠尿液影响的代谢组学初步研究

【摘要】 目的:利用基于HPLC/MS/MS的代谢组学分析方法研究龙胆给药大鼠尿液的代谢物变化,寻找可能的生物标记物。方法:SD大鼠灌胃给予14 g·kg-1龙胆水提液,连续33天,尿样经SPE处理,进行HPLC/MS/MS分析,利用PCA分析数据,从中找出生物标记物。结果:给药33天,龙胆组与对照组大鼠尿样代谢组有较大差异,尤其是雄性大鼠更为明显。生物标记物可能是牛磺酸、甲状腺素、苹果酸等物质,这与龙胆保肝、利胆、抗甲亢、影响能量代谢的药理作用相一致,同时发现性别差异也是造成代谢组变化的原因之一。结论:长期给予寒性中药会对实验动物的代谢产生影响,为进一步阐释吴茱萸的药性研究工作提供依据。
【Abstract】 AIM:HPLC/MS/MS-based metabonomics research on the change of endogenous metabolites of Sprague-Dawley Rats administrated of aqueous extract of Radix Gentianae. METHODS:Sprague-Dawley rats successively administrated 14g·kg-1 aqueous extract of Radix Gentianae for 33 days,urine were collected by metabolism cages and detected by using the HPLC/MS/MS.All dates were analyzed by the principal component analysis(PCA) using the SIMCA-P 10. 0 software.RESULTS:The PCA demonstrated that the metabolome between administrated group and control group had difference in rats urine sample after administrated for 33 days,especially in male rats,dosed group had significant changes in biomarkers compared with control group which possibly were taurine,thyroxine,malic acid and other compounds, the result was consistent with pharmacological effects of Radix Gentianae,such as liver protection,cholagogue, anti-hyperthyroidism,changing energy metabolism, the difference of sex was also found.CONCLUSION:The Gentianae had changed the endogenous metabolites of Sprague -Dawley Rats.This work can provide base for the further research on the interpretation of drug property of evodiae.
