放置时间对布洛芬栓生物利用度影响的研究

【摘要】 目的:建立布洛芬血药浓度的测定方法,考察放置时间对布洛芬栓生物利用度的影响,并分析影响原因。方法:以乙腈:20 mmol·L-1磷酸二氢钾=64:36(v/v)为流动相,样品蛋白沉淀后直接进样。20名健康男性受试者单剂量、交叉直肠给予150 mg新制布洛芬栓(A)及放置1.5年的布洛芬栓(B)后,采用上述方法测定人体血浆中不同时间点布洛芬的浓度,计算其药代动力学参数和相对生物利用度,评价放置时间对布洛芬栓生物利用度的影响。结果:血浆中布洛芬与内源性杂质分离完全,在0.3125 mg·L-1~20.0 mg·L-1浓度范围内与峰面积比线性良好,最低定量浓度为0.3125 mg·L-1,方法学验证符合生物样本分析要求。单次给予150 mg制剂A或制剂B后的AUC0-8分别为(22.1±5.7)、(15.5±4.0)mg·h·L-1,Cmax分别为(6.3±1.8)、(3.3±1.2)mg·L-1,Tmax分别为(1.5±0.4)、(4.2±1.2)h,t1/2分别为(2.1±0.8)、(4.2±2.9)h。方差分析结果表明新制布洛芬栓的AUC0-8,Cmax及生物利用度均高于放置1.5年布洛芬栓。制剂A相对于制剂B的生物利用度为(153±62)%。结论:布洛芬栓剂的吸收速度和吸收程度与储存期有关。严格的储存条件是保证该制剂的质量关键。
【Abstract】 AIM:An HPLC method was developed and validated to determine ibuprofen in plasma and study the influence of time of storage to bioavailability of ibuprofen suppository. METHODS:Ibuprofen was detected after proteinprecipitation with a mixture of acetonitrile-phosphate buffer solution(64/36 v/v) as the mobile phase.A single dose of 150mg ibuprofen suppository prepared freshly(A) and stored for 1.5 years(B) was administered to 20 healthy volunteers in a randomized crossover study,and the pharmacokinetics and bioavailability were studied.RESULTS:ibuprofen was separated well with the endogenous substances,the calibration curve was linear in the range of 0.3125-20.0 mg·L-1 with r2=0.9996.The AUC0~8 of the two suppositories were (22.1±5.7) and(15.5±4.0)mg·h·L-1,Cmax were(6.3±1.8) and(3.3±1.2) mg·L-1,Tmax were(1.5±0.4) and(4.2±1.2) h,T1/2 were(2.1±0.8) and(4.2±2.9) h. the relative bioavailability of suppository A was(153±62) %when compared with suppository B.CONCLUSION:Statistic analysis showed significant difference between the two suppositories,and the AUC0-8,Cmax,relative bioavailability of suppository A was higher than B.The established method is accuracy and sensible for determination ibuprofen in human plasma.
