多次口服奥美沙坦片在健康人体的药代动力学

【摘要】 目的:研究多次口服奥美沙坦片在中国健康受试者体内的药代动力学。方法:10名受试者第1天和第8～14天,每天口服20 mg奥美沙坦片。采用HPLC-MS法测定给药后不同时间奥美沙坦的血药浓度。利用DAS Ver 2.0和SPSS13.0计算药动学参数和进行统计分析。结果:单次和多次口服20mg.奥美沙坦片达稳后,主要药代动力学参数:AUC0→t分别为（3356.3±865.9）、（4764.7±1700.5）ng·h/mL;AUC0→∞为（3778.2±922.9）、（4987.4±1679.7）ng·h/mL;Cmax为（295.4±70.1）、（550.0±157.3）ng/mL;tmax为（2.4±0.7）、（2.3±0.8）h;t1/2为（14.9±5.0）、（12.4±4.8）h。结论:中国健康受试者连续口服20 mg奥美沙坦片体内未见蓄积。
【Abstract】 AIM:To study the pharmacokinetics of olmesartan tablets after multiple oral dose adiminstration in Chinese healthy volunteers. METHODS:10 healthy volunteers were randomized to administer a single dose of olmesartan tablets at 20 mg on day 1 and day 8 to day 14.The concentrations of olmesartan in human plasma were determined by HPLC-MS.The main pharmaockinetic parameters were calculated and analyzed by DAS ver 2.0 and SPSS 13.0.RESULTS: The main pharmacokinetic parameters of olmesartan after oral a single dose and multiple-dose were as follow:AUC0→t（3356.3±865.9） and （4764.7±1700.5） ng·h/mL;AUC0→∞（3778.2±922.9） and （4987.4±1679.7） ng·h/mL;Cmax（295.4±70.1） and（550.0±157.3） ng/mL;tmax（2.4±0.7） and（2.3±0.8） h;t1/2（14.9±5.0） and（12.4±4.8） h,respectively.CONCLUSION:There is not cumulation of olmesartan in plasma after multiple dose of 20 mg.
