靛玉红甲肟对人食管癌细胞系体外增殖和细胞周期的影响

【摘要】 目的:探讨靛玉红甲肟(Indirubin-3’-monoxime)对体外培养的人食管癌细胞系EC-1和Kyse70增殖和细胞周期的影响。方法:不同浓度靛玉红甲肟处理培养的食管癌EC-1和Kyse70细胞后,采用MTT法检测细胞的增殖活性,流式细胞仪和RT-PCR检测细胞周期分布及bcl-2和bax mRNA的变化。结果:靛玉红甲肟对人食管癌细胞EC-1和Kyse70的生长具有明显的抑制作用,其作用表现为剂量依赖性和时间依赖性(P<0.05)。流式细胞仪检测发现:以5μmol/L,的靛玉红甲肟对EC-1细胞处理24、48、72 h后,G0/G1期细胞比例逐渐下降(P<0.05),S期细胞比例未见明显改变,G2/M期细胞比例逐渐上升(P<0.05),呈时间依赖性。RT-PCR检测发现人食管癌细胞bcl-2和bax mRNA比例呈时间依赖性下调。结论:靛玉红甲肟对人食管癌细胞系EC-1和Kyse70具有明显的增殖抑制作用,其作用机制与阻滞细胞周期G2/M期和下调bcl-2/bax表达比例有关。
【Abstract】 Objective:To observe the effect of Indirubin-3’-monoxime on proliferation and cell cycle of human esophageal cancer cell line EC-1 and Kyse70.Methods:EC-1 and Kyse70 cells were treated with Indirubin-3’-monoxime.The proliferative status of cells was measured by methabenzthiazuron(MTT) assay.Cell cycle and expression of bcl-2,bax mRNA was analyzed, detected by flow cytometry(FCM) and RT-PCR.Results:Indirubin-3’-monoxime inhibited growth of EC-1 and Kyse70 cells in a dose-dependent and time-dependent manner(P<0.05).Cell G0/G1 phase ratio decreased gradually(P<0.05),S-phase ratio has not changed significantly,G2/M phase ratio increased gradually after exposed to Indirubin-3’-monoxime 24、48、72h in a dose dependent manner(5μmol/L)(P<0.05),RT-PCR detection of esophageal cancer cells were found in bcl-2 and bax mRNA ratio down in a dose dependent manner(5μmol/L)(P<0.05).Conclusions: Indirubin-3’-monoxime inhibits the proliferation in human esophageal cancer line EC-1 and Kyse70.The effect mechanism is related to arrest the G2/M phase in cell cycle and bcl-2 and bax mRNA ratio down.
