肉桂酸、邻菲罗啉稀土配合物的合成、表征及其与DNA的作用

【摘要】 本文研究了稀土配合物与DNA的作用方式。分别合成了以Pr、Nd、Ho、Tm、Yb、Lu为中心,以肉桂酸(Hcin)和邻菲罗啉(Phen)为配体的六种三元稀土固态配合物。经C、N、H元素分析和稀土络合滴定确定了三元配合物的组成为RE(cin)3phen(RE3+=Pr3+、Nd3+、Ho3+、Tm3+、Yb3+、Lu3+)。测定了配合物的红外光谱及紫外光谱,结果表明配合物中的RE3+离子与肉桂酸中羧基的氧原子及邻菲罗啉中的两个氮原子配位成键。采用紫外光谱法研究了稀土三元配合物与小牛胸腺DNA的作用,随着DNA浓度的增加,配合物的最大吸收峰发生红移,且最大吸收峰处的吸光度逐渐减小,表明稀土配合物以插入的方式和小牛胸腺DNA发生了强烈的作用。采用荧光光谱法以不同浓度配合物与EB-DNA体系相互作用,随着配合物浓度的增加,EB-DNA体系的荧光强度明显减弱,出现了荧光猝灭现象,说明配合物与EB以竞争方式结合DNA,进一步证明了配合物以插入的方式与DNA作用。
【Abstract】 Interaction of rare earth complexes and DNA was studied in this paper.Six complexes of rare earth(RE3+=Pr3+、Nd3+、Ho3+、Tm3+、Yb3+、Lu3+)were synthesized with Cinnamic acid(Hcin)as the first ligand and 1,10-Phenanthroline as the second ligand.The chemical formula of the complexes was RE(cin)3phen(RE3+=Pr3+、Nd3+、Ho3+、Tm3+、Yb3+、Lu3+)characterized by elemental analysis and rare earth coordination titration.IR spectra and UV spectra have been studied and the results indicated that the rare earth ions are bounded with the carboxyl oxygen atoms of Cinnamic acid,and with the N atoms of I, 10-Phenanthroline.The actions of the rare earth complexes and CT-DNA with UV spectra were researched. When the concentration of CT-DNA is increased,the largest absorption peak is gradually Red shift and also weak,which showed that the interaction modes of complexes and CT-DNA are intercalation.Fluorescence quench experiment was also used to study the interaction between the complexes and ethidium bromide-DNA(EB-DNA)system.As the concentration of the complexes is increased,fluorescence intensity is gradual weaken,which also proved that the interaction mode between the complexes and DNA is intercalation.
