鸡NCOA7基因在成肌细胞分化和腿肌组织发育中的表达特性分析

【摘要】 NCOA7(Nuclear receptor coactivator 7)属于雌激素受体蛋白。在前期采用Illumina60k SNP芯片开展的胴体性状的全基因组关联(GWAS)研究中发现,NCOA7基因与胸肌、腿肌重达到显著相关。本试验采用Q-PCR技术,研究鸡NCOA7基因组织表达特性、成肌细胞增殖与分化期以及腿肌组织中的发育过程表达规律。结果显示:NCOA7在多个组织中均有表达,在腿肌中的表达量显著高于其他组织(P<0.05)。对成肌细胞的研究发现,NCOA7基因在增殖期的成肌细胞中的表达量显著低于分化晚期(P<0.05)。在成肌细胞分化过程中,NCOA7基因的表达量随着诱导分化时间的延长而逐漸升高,96 h达到顶峰。在胚胎发育后期腿肌中的发育模式为:随着孵化时间的延长,NCOA7的表达量逐渐升高。出生后至84日龄的表达量表现为逐渐降低。结论,初步推断NCOA7基因参与了肌肉组织发育的调控。
【Abstract】 NC0A7(Nuclear receptor coactivator 7) belongs to the estrogen receptor protein.In our previous study, the results of genome-wide association studies(GWAS) on the carcass traits using Illumina60k SNP chip revealed that the NCOA7 gene was significantly correlated to breast and leg muscle weight.mRNA expression of NCC’A7 gene levels of 10 tissues were compared.And the variation of mRNA expression of NC0A7 gene in muscle tissue? including the proliferation and differentiation of myoblasts and also the late embryonic period(14~21 days) to 84th cays after hatch was identified by Q-PCR.Results showed that NCOA7 expressions in the leg muscle was significantly higher than other tissues(P<0.05).From the expressions in proliferation and differentiation of myoblasts analysis,it was ol served that the mRNA expression level of NCOA7 gene in the differentiation was significantly higher than the proliferation(P<0.05), and increased with the differentiation process,and peaked at 96 h.The relative mRNA expression level of breast muscle in embryonic period showed that NC0A7 mRNA expression levels were continued to rise during el4d to e21d where el4d to el8d showed comparatively faster.The expression of development stages from hatch to 84th day was reduced gradually.Results of various analysis showed that NC0A7 gene was involved in the regulation of muscle development.
