中国环颈雉脂蛋白酯酶基因cDNA序列测定及组织表达分析

【摘要】 运用RT-PCR方法从中国环颈雉的总RNA中克隆了脂蛋白酯酶(LPL)基因的cDNA部分序列,然后运用Real-time PCR法检测了LPL基因在中国环颈雉不同发育阶段、不同组织及不同品种间的表达情况。结果表明,中国环颈雉LPL基因cDNA序列为791 bp,与鸡的基因序列同源性较高。中国环颈雉LPL基因在150日龄表达量最大,胸肌为优势表达组织,与活重、胸肌和腿肌肌內脂肪含量存在相关性。中国环颈雉LPL基因的研究,为进一步研究中国环颈雉的生长调控和育种繁育提供理论依据。

【Abstract】 Lipoprotein lipase(LPL) cDNA was cloned from Chinese ring-necked pheasant using RT-PCR method, and then LPL gene expression was detected using real-time PCR method in different developmental stages,different tissues and different breeds.The results were as follows:The cloned cDNA fragment was 791 bp,the nucleotide and amino acid sequences of Chinese ring-necked pheasant LPL had the closest phylogenetic relationship with chickens.Expression of LPL gene was the highest when Chinese ring-neck pheasant was 150 days of age.LPL gene could express highly in the breast muscle.There were correlations among LPL gene expression and live weight,breast and leg muscle intramuscular fat content.LPL gene was studied to provide theoretical basis for regulation of growth and genetic breeding in Chinese ring-necked pheasant.
