一株硝化反硝化菌Pseudomonas putida F6的筛选鉴定及反硝化特性研究

【摘要】 为了筛选出高效脱氮益生菌应用于改善养殖水体水质,本试验通过酸碱指示剂筛选等方法,从草鱼养殖池塘中分离筛选出了一株具有硝化和反硝化功能的菌株。在以氯化铵为氮源时,该菌株培养24h对氨氮的去除率达91.44%,以亚硝酸钠和硝酸钠为氮源,对亚硝酸氮和硝酸氮的去除率分别达37.79%和63.37%。其16S rDNA序列与恶臭假单胞菌(Pseudomonas putida)的同源性达98%,命名该菌为Pseudomonas putida F6。反硝化特性研究表明:在好氧条件下,该菌分别以亚硝酸盐和硝酸盐为氮源都有较好的生长和脱氮能力,其对亚硝酸盐氮和硝酸盐氮去除率最高分别可达100%和80.77%,脱氮率最高分别可达78.75%和82.37%(脱氮率以TIN的去除率表示)。同时发现,该菌培养12h之前生长较快,脱氮效果明显。该菌适宜的生长条件:亚硝酸氮和硝酸氮浓度分别为10-60mg/L和10-40mg/L,pH、温度和摇床转速均为6-9、25-30℃和50-200rpm;脱氮的适宜条件为:氮浓度10mg/L、pH 6-9、20-30℃、摇床转速150rpm(溶氧5.1mg/L)。该菌在模拟草鱼池水中的稳定性较好,8天后的存活率为46.5%。

【Abstract】 To screen bacteria with high nitrogen removal rate for improving water quality,a denitrifier F6 with nitrification characteristics was isolated from a grass carp pond and was identified as Pseudomonas putida by 16 S rRNA sequencing.Study of the denitrification characteristics showed that:the removal rate of NH4--N,NO2--N and NO3--N reached 91.44%,37.79% and 63.37% respectively.The strain had optimal growth and denitrification rate in the culture medium using NO2--N or NO3--N as the sole nitrogen source.Highest removal rate for NO2--N and NO3--N reached 100% and 80.77%,respectively,and highest removal rate for total inorganic nitrogen reached 78.75% and 82.37%,respectively.Its logarithmic phase was from 4 to 24h,and denitrification mainly occurred during the 4-12h phase.The best growth of strain F6 occured at a concentration of 10-60mg/L NO2—N and 10-40mg/L NO3--N,pH6-9,25-30℃ and 50-200rpm shaking speed.The best denitrification condition for F6 was as follows:10 mg/L nitrogen,pH 6.0-9.0,20-30℃ and 150rpm(about DO 5.1mg/L).The strain was relatively stable in the water of grass carp pond,46.5% of which can survive after 8 days cultivation.
