乳酸球菌传递的pEGF对早期断奶仔猪小肠黏膜形态和二糖酶活性的影响

【摘要】 本文旨在研究表达EGF的乳酸菌对早期断奶仔猪小肠的组织形态与肠黏膜二糖酶活性的影响。实验选取21日龄体重相近的仔猪108头分4个处理组,每组3个重复,每个重复9头,4个处理组按2×2分组设计,即高质量+EGF组(HE)、高质量组(HM)、低质量+EGF组(LE)和低质量组(LM)四个处理。HE、LE两组则从断奶的当天开始每日饮用含基因工程表达的猪EGF(pEGF)及其转基因乳酸球菌(EGF-Lactococcus lactis,EGF-LL)的M17培养液,HM、LM两组则仅给予同样剂量的M17培养液。在试验的第15天,从每个处理中随机抽取6头(共24头)试猪屠宰测定其肠绒毛高度、腺窝深度和肠黏膜二糖酶的活性。结果表明:LE组的空肠绒毛高度(V)、腺窝深度(C)及绒毛高度/腺窝深度(V/C)均极显著地大于LM组(P<0.01),HE组的腺窝深度和绒毛高度/腺窝深度显著大于HM组:LE和HE组空肠处的乳糖酶和蔗糖酶活性均显著高于LM组(P<0.01),十二指肠处HE组乳糖酶活性极显著地高于LM组(P<0.01),空肠处LE组的乳糖酶活性显著高于HM组(P<0.05),LE组的小肠黏膜形态和二糖酶活性与HM组相差不大。结论:口服表达EGF的乳酸菌可以改善断奶仔猪小肠形态,提高消化酶的活性,对空肠的影响尤为显著,其效果和血浆蛋白粉的相差不大。

【Abstract】 The experiment was to study the effects of EGF-expressing L.lactis(EGF-LL)on intestinal morphology and disaccharidases activities in the early weaned pigs.108 weanling piglets at 21d of age were randomly allocated into 4 treatments.Each treatment was assigned 3 replicates of 9 pigs each.They were named as high quality feed+EGF(HE)、high quality feed+M17(HM)、low quality feed+EGF(LE)、low quality feed+M17(LM).The HE and LE groups were fed with M17 contained EGF-expressing L. lactis(EGF-LL) from the day of the weaned;the HM and LM groups were given the same doses of M17 culture solution only.After the treatments for 14d,six pigs randomly selected from each treatment were killed to examine the mean villus height,crypt depth and intestinal disaccharide activity.In the jejunum, the villus height(V),crypt depth(C) and V/C of LE group were significantly higher than LM group (P<0.01),the crypt depth and V/C of HE group were higher than HM group;In the jejunum,the activities of lactase and sucrase of LE and HE group were both higher than LM group(P<0.01),the activity of lactase of LE group was significantly higher than LM group(P<0.05),in the duodenum,the activity of lactase of HE group was significantly higher than LM group(P<0.01),intestinal morphology and disaccharidase activity of LE group is no significant difference with HM group.The results indicate that oral administration of EGF-LL could improve the small intestinal morphology and disaccharidases activities of the early weaned pigs,especially to the jejunum.The effect of the EGF is no significant difference with the plasma protein powder.
