GnRH在广西摩拉杂交水牛乳腺的免疫组化定位及其受体基因表达的研究

【摘要】 为了探索促性腺激素释放激素(GnRH)在广西摩拉杂交水牛乳腺的分布特点,本研究采用免疫组化及RT-PCR技术检测广西摩拉杂交水牛乳腺组织中的促性腺激素释放激素(GnRH)及其受体。结果表明,15份正常乳腺组织中有13份呈GnRH免疫阳性反应,免疫阳性产物主要集中在腺泡细胞的胞质和胞膜,胞核无特异性染色;同时扩增的目的条带大小约1008bp,与促性腺激素释放激素受体(GnRHR)基因片段大小一致。为深入阐明GnRH调节乳腺功能的机制提供理论依据。
【Abstract】 In order to study the distribution of Gonadotropin-Releasing Hormone In in the mammary gland of Guangxi hybrid Murrah Buffalo.we used immuno-histochemical and reverse transcription PCR to detected gonadotropic releasing hormone(GnRH ) and gonadotropic releasing hormone receptor(GnRH-R ) in the mammary gland of Guangxi hybrid Murrah Buffalo.The result show that 13 pieces tissue were positive in 15 pieces normal mammary gland tissue,the positive substance were mainly distributed in periplast and membrane of acinous cell and it was negative in nucelus;Meanwhile, the GnRH -R gene fragment was amplified with 1008 bp in size.The study paved the way for elucidating the mechanism of mammary gland function regulation by GnRH.
