高能低蛋白日粮中添加甜菜碱对蛋鸡肝脏apo AⅠ和apo B100基因mRNA表达的影响

【摘要】 (目的)本试验以海蓝褐蛋鸡为试验对象,研究高能低蛋白日粮中添加甜菜碱对蛋鸡肝脏中apo AI和apoB100基因mRNA表达的影响。(方法)选300日龄海蓝褐蛋鸡135羽,随机分成3组(每组设3个重复,每个重复15羽),分别饲喂正常日粮(对照组,代谢能11.30MJ/kg,粗蛋白17.20%)、高能低蛋白日粮(高能低蛋白组,代谢能12.81MJ/kg,粗蛋白13.04%)、添加1000 mg/kg甜菜碱的高能低蛋白日粮(甜菜碱组,代谢能12.81MJ/kg,粗蛋白13.04%),正试期60 d,分别于试验1 d、30 d、60 d,每组选取体重相近的试验蛋鸡6羽,取肝脏,采用实时荧光定量PCR方法测定肝脏中apo A I和apo B100基因mRNA的相对表达量。(结果)与对照组相比,饲喂高能低蛋白日粮30天可导致蛋鸡肝脏中apo A I的mRNA表达显著降低(P<0.05),apo B100的mRNA表达显著升高(P<0.05),饲喂60天可导致蛋鸡肝脏中apo A I和apo B100的mRNA表达均极显著降低(P<0.01);与同期高能低蛋白组相比,添加1000mg/kg甜菜碱可使蛋鸡肝脏中apo A I和apo B100 mRNA的表达水平均升高,差异显著(P<0.05)。(结论)结果提示,高能低蛋白日粮中添加甜菜碱可通过提高肝脏中apo A l和apo B100 mRNA的表达,增强肝脏中脂类物质的转运出。
【Abstract】 (Objective) The experiment was conducted to investigate the influence of high-energy low-protein diet supplemented with betaine on apo AⅠand apo B100 mRN A expression in liver of laying hens.(Method) One hundred and thirty five birds(Hy-line Variety Brown laying hens,Gallus domesticus) aged 300 days were randomly allotted into 3 dietary treatments of 45 broilers each.The groups were:1) Control group(CG,semisynthetic commercial layer standard diet in accordance with the nutrient requirements of poultry(National Research Councile,1998),ME 11.30MJ/kg and CP 17.20%); 2) high energy-low protein group(HELPG,high energy-low protein diet which were partially formulated in accordance with Controls,ME 12.81 MJ/kg and CP 13.04%);3) betaine group(BG,the high energy-low protein diet supplemented with 1000 mg betaine/kg DM,ME 12.81 MJ/kg and CP 13.04%).All birds were reared for 60 days under normal condition in the Jiangxi Agriculture University Poultry form and were fasted overnight with a free access to water before sacrificed.Apo A I and apo B100 mRNA expression in liver of six layers(stratified by weight,1.6-1.7 kg ) from each group were recorded by real-time fluorescent quantitative PCR on day of 1st,30th and 60th.(Result) The results showed that compared with the CG, the level of apo A I mRNA expression in liver of layers fed with high energy-low protein diet decreased significantly on the day of 30(P<0.05),while the level of apo B100 mRNA expression increased significantly(P<0.05).After 60 days fed with the high energy-low protein diet,the level of apo A I and apo B100 mRNA expression were decreased very significantly (P<0.01).Compared with the same period of HELPG,the high energy-low protein diet supplemented with 1000 mg betaine/kg DM could result in the increasing significantly of the levels of apo A I and apo B100 mRNA expression in liver of layers(P<0.05).(Conclusion) These results indicated that betaine could protect the laying hens from fatty liver induced by high energy-low protein diet through increasing the expression levels of liver apo A I and apo B100 mRNA,and thus enhancing the transportation of lipids from the liver.
