1α,25-(OH)2D3通过成骨细胞M-CSF途径影响破骨细胞增殖与分化

【摘要】 【目的】探讨不同浓度1α,25-(OH)2D3(0、10-9、10-8、10-7mol/L)对体外培养SD大鼠成骨细胞(osteoblasts,OB)内巨噬细胞集落刺激因子(M-CSF)mRNA表达以及M-CSF对破骨细胞(osteoclasts,OC)增殖与分化的影响。【方法】lα,25-(OH)2D3与OB共培养24、48、72 h,采用FQ-PCR法测定M-CSF mRNA表达量。M-CSF与骨髓单核细胞共培养9 d,抗酒石酸酸性磷酸酶(TRAP)染色观察并计数OC量,扫描电镜观察象牙片吸收陷窝。【结果】与对照组比较,不周浓度1α,25-(OH)2D3作用48、72 h均显著(P<0.05)或极显著(p<0.01)促进OB内M-CSF mRNA的表达;同时,M-CSF较对照组促进骨髓单核细胞贴壁与增殖,并能与RANKL联合诱导OC的形成与活化。【结论】1α,25-(OH)2D3能上调OB内M-CSFmRNA的表达,间接促进OC的生成和活化,增强骨更新。

【Abstract】 [Objective]To investigate the expression of M-CSF mRNA,osteobalsts obtained from Sprague Dawley rat were treated with different concentrations of 1α.25-(OH)2D3(0,10-9,10-8,10-7 mol/L).Meanwhile,to investigate the effects of M-CSF on osteoclasts’ proliferation and differentiation,bone marrow mononuclear cells(BMM) were cultured with or without M-CSF. [Method]The expression of M-CSF mRNA were determined through FQ-PCR.The formation of OC were detected by histochemical staining for tartrate-resistant acid phosphatase(TRAP),and the detection of lacunar resorption via scanning electron microscopy at the 9th day.[Result]Compared with the control group,different conentrations of 1α,25-(OH)2D3 can significantly or very significantly induce the expression of M-CSF mRNA(P< 0.05 or P< 0.01).M-CSF can promote BMM adherence and proliferation,and it can induce formation and activation of osteoclasts combined with RANKL.[Conclusion]1α,25-(OH)2D3 can enhance bone turnover through facilitating the generation and activity of osteoclasts via up-regulation of M-CSF mRNA.
