【摘要】 采集16头广西本地沼泽型水牛瘤胃内容物,提取总DNA,利用产甲烷古菌通用引物扩增I6SrRNA基因序列,进行DGGE分析。将DGGE凝胶上7条优势条带进行回收克隆测序,得到9条序列,其中3条与甲烷短杆菌(Methanobrevibacter)的同源性也高达99%。1条与甲烷短杆菌的同源性达98%,1条序列与广古菌门(Euryarchaeota)热原体目(Thermoplasmatales)的同源性达98%。另外,有4条是和古菌存在序列相似性的未培养细菌,分别属于拟杆菌(Bacteroidetes),厚壁菌(Firmicutes),变形菌门(Proteobacteria.),几个细菌门类之间无太大同源性。更多还原

【Abstract】 collected rumen contents from sixteen local swamp Buffalo in Guangxi,extracted total DNA,used the universal primers amplified methanogenic archaea hypervariable region of 16S rRNA gene sequences,and did DGGE analysis. Seven superiority bands were carriy on clone and then nine sequences were obtained.There were three sequences had homology with Methanobrevibacter as high as 99%,one sequence had homology with Methanobrevibacter was 98%,and one sequence unclassified in Euryarchaeota had 98%homology with Thermoplasmatales.In addition,four sequences which were similar with methanogenic archaea were uncultured bacteria,belonging to Bacteroidetes Firmicutes and Proteobacteria.But these bacterium classes does not have the big homology更多还原
