枯草芽孢杆菌B1对高脂日粮诱导小鼠氧化应激的保护作用
Protection of Bacillus subtilis B1 against oxidative stress of rat fed with high-fat diet

【摘要】 将45只ICR雄性小鼠,随机均分为三组,每组3个重复,每个重复5只:A组为空白对照组,饲喂小鼠基础日粮;B组为氧化应激组,饲喂高脂日粮;C组为益生菌组,在高脂日粮中添加108CFU/g枯草芽孢杆菌B1制剂,试验期为30d。试验结束后测定了各处理组小鼠增重、血脂等血清生化指标及血清抗氧化功能。结果表明:高脂日粮显著增加了小鼠的末期体重(8.57%,P<0.01)和日增重(58.33%,P<0.05),而芽孢杆菌B1则显著降低了高脂日粮引起的小鼠的增重(P<0.01)。血清生化指标测定表明,高脂日粮显著升高了小鼠血清中的总胆固醇(Total cholesterol,TC)含量、葡萄糖浓度和谷丙转氨酶(Glutamic-pyruvic transaminase,GPT)活性,并显著降低了血清抗O2·-活力、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)活性及还原型谷胱甘肽(Reducedglutathione,GSH,P<0.01)含量,而添加芽孢杆菌B1则使小鼠血清中葡萄糖浓度、尿素氮含量、谷草转氨酶(Glutamic-oxalacetic transaminase,GOT)和GPT活性均显著降低(P<0.05),并显著提高了小鼠血清总抗氧化能力(P<0.05)。以上结果提示枯草芽孢杆菌B1能够有效地提高小鼠血清抗氧化活性、保护小鼠机体免受高脂日粮对小鼠的损伤,并减轻高脂日粮引起的小鼠肥胖。
【Abstract】 To investigated the protection effect of Bacillus subtilis B1(B.subtilis B1) against oxidative stress of rat fed with high-fat diet,45 male ICR mice were randomly divided into three groups(Group A-C,three replicates for each group): Group A(Control group) was fed with basal diet;Group B(Oxidative stress group) was fed with high-fat diet;Group C (Probiotics group) was fed with high-fat diet,but supplemented with Bacillus B1(10~8CFU/g).The trial was lasted for 30d. The rat weight gain,serum biochemical index including blood fat,and serum antioxidant capacity were analyzed at the end of the experiment.The results showed that high fat diet significantly increased the rat’s final weight(8.57%,P<0.01) and daily weight gain(58.33%,P<0.05).B.subtilis B1 significantly decreased the rats weight gain caused by high fat diet (P<0.01).Serum index analyze showed that fat diet significantly increased rat serum TC level,glucose concentration and GPT activity,and significantly decreased serum anti-superoxide anion capacity,GSH-Px activity and GSH level(P<0.01). Supplementation of B.subtilis B1 significantly decreased serum glucose concentration,BUN level,and GOT and GPT activities(P<0.05) and increased serum antioxidative activities(P<0.05).These results indicated that B.bacillus B1 could efficiently improve rat serum antioxidant capacity,protect rat against inordinate weight gain and injury of oxidative stress caused by high-fat diet.
