流式细胞仪分离精子过程对水牛精子顶体的影响

【摘要】 本实验的目的是检测用流式细胞仪分离水牛精子过程中的各个阶段对精子顶体损伤情况及公牛个体间的差异。主要采用考马斯亮蓝染色法(CBB),依次对鲜精组(fresh)、染色组(stained)、分离组(sexed)和冷冻-解冻组(frozen-thawed)四个阶段的精子进行检测。结果表明,每个阶段顶体损伤情况各异,并且顶体损伤程度从鲜精组至冷冻-解冻组逐渐增大,但是染色组分别与鲜精组和分离组之间差异不显著(P>0.05),而分离组顶体损伤显著大于鲜精组(P<0.05)。冷冻解冻组顶体损伤远远大于前三个组(P<0.05)。结论:染色过程对精子顶体造成了一定的损伤,但损伤不大,而分离和冷冻-解冻对精子顶体造成极大地损伤。不同阶段对顶体的损伤存在公牛个体差异。

【Abstract】 The objective of this study was to detect the acrosome damage of buffalo separated sperm sorting by Flow Cytometry at different stages.Buffalo sperm at four stages(fresh,stained,sorted,frozen-thawed) were measured by Coomassie Brilliant Blue R250(CBB) stain.The result showed that the degree of acrosome damage of sperm during sorting process varied from stage to stage and the rate of sperm with acrosome damage was increased from fresh to frozen-thawed stages.There was no significant differences(p>0.05) between stained and fresh groups.However,the acrosome of the sorted sperm was much more damaged(p<0.05) than the fresh sperm. The degree of acrosome damage with frozen-thawed group was the highest when compared with the other three groups(P<0.05).In conclusion,staining could be harmful to buffalo sperm acrosome,but the impact is not serious; in addition,the sorted and frozen-thawed process could do great damage to the buffalo sperm and the different degrees of such a acrosome damage is existed with individual buffalo bull.
