【摘要】 新生牛睾丸解离成单细胞悬液,差速贴壁法纯化支持细胞后进行体外培养与鉴定。结果表明:2%血清的培养液能够用于支持细胞的体外培养;支持细胞体外培养能够传至第20代,随着传代次数的增加,增值速度越来越慢;免疫组化染色显示,波形蛋白呈阳性,RT-PCR分析显示,表达支持细胞的标志基因SCF和GDNF,第15代的支持细胞含有正常的二倍体核型。更多还原

【Abstract】 Testes were digested into single cell suspension which was isolated and purified. Purified cells were in vitro culture and identificated.The results were showed as following: DMEM/F12 medium supplemented with 2%serum could be used to culture Sertoli cells,Sertoli cells in vitro could be passaged to the 20th generation,with the increase in the number of passages, the proliferative rate became slower;vimentin examination was positive in Sertoli cells,RT-PCR analysis showed that expression of marker genes in Sertoli cells of the SCF and GDNF,and the 15th generation of the cells maintained a normal diploid karyotype.更多还原
